长柄扁桃正丁醇提取物降血脂作用的量效关系研究

摘要 目的:探索长柄扁桃降血脂正丁醇提取物降血脂作用的量效关系。方法:大鼠随机分成正常对照组和高脂模型组,5周后将成模大鼠随机分为高脂模型对照组、血脂康阳性药组、长柄扁桃正丁醇提取物0.75 g/kg、1.50 g/kg、3.00 g/kg和4.00 g/kg剂量组。给药5周后,观测大鼠体重、血清中胆固醇(total cholesterol,TC)、甘油三酯(triglyceride,TG)、高密度脂蛋白(high density lipoprotein cholesterol,HDL-C)、低密度脂蛋白(low density lipoprotein cholesterol,LDL-C)、丙二醛(malondialdehyde,MDA)、超氧化物歧化酶(superoxide dismutase,SOD)含量及谷丙转氨酶(alanine aminotransferase,ALT)、谷草转氨酶(aspartate aminotransferase,AST)活性并计算肝脏系数、动脉硬化指数(arteriosclerosis index,AI)、HDL-C/TC和SOD/MDA。结果:长柄扁桃1.50 g/kg和3.00 g/kg剂量组均可降低大鼠血清中TC、TG、LDL-C、MDA、ALT、AST、肝系数及AI值(P<0.05),提高SOD活性、SOD/MDA和HDL-C/TC值(P<0.05)。结论:长柄扁桃正丁醇提取物降血脂作用有效剂量范围为1.50~3.00 g/kg之间。 Objective:To explore the dose-effect relationship of the blood lipid-lowering effect of n-butanol extract of Amygdalus pedunculata Pall.Methods:The rats were randomly divided into normal control group and high-fat model group.After 5 weeks,the rats that had formed a hyperlipidemia model were randomly divided into a hyperlipidemia model control group,a Xuezhikang group,and a Amygdalus pedunculata Pall n-butanol extract 0.75 g/kg,1.50 g/kg,3.00 g/kg and 4.00 g/kg dose groups.After 5 weeks of treatment,we observe the rat body weight,serum total cholesterol(TC),triglyceride(TG),high density lipoprotein cholesterol(HDL-C),low density lipoprotein(LDL-C),malondialdehyde(MDA),superoxide dismutase(SOD)content,alanine aminotransferase(ALT),aspartate aminotransferase(AST)Active and calculate liver coefficient,arteriosclerosis index(AI),HDL-C/TC and SOD/MDA.Results:Both the 1.50 g/kg and 3.00 g/kg dose groups of the n-butanol extract of Amygdalus pedunculata Pall can significantly reduce the serum TC,TG,LDL-C,MDA,ALT,AST,and liver coefficients in hyperlipidemia rats and AI value(P<0.05),can significantly improve SOD,SOD/MDA and HDL-C/TC values(P<0.05).Conclusion:The effective dose range of the n-butanol extract of Amygdalus pedunculata on the hypolipidemic effect of hyperlipidemia rats is 1.50~3.00 g/kg.
