家蚕卵中间感温过程中谷胱甘肽氧化还原代谢的变化

摘要 家蚕越年蚕种的初春中间感温是复式冷藏法的一个技术环节,而谷胱甘肽氧化还原代谢与家蚕胚胎滞育诱导和解除密切相关。为了解中间感温期间蚕卵中谷胱甘肽氧化还原代谢的生理生化变化,以菁松×皓月(JS×HY)杂交种为材料,对其初春中间感温过程中的谷胱甘肽含量与谷胱甘肽氧化还原循环相关酶活性进行测定和比较分析。结果发现:GSH(还原型谷胱甘肽)相关酶活性变化对抵抗温度伤害有一定作用,蚕卵解除滞育需要高GR(谷胱甘肽还原酶)活性,GR活性显著降低是GSH/GSSG(氧化型谷胱甘肽)比值显著降低的主要原因之一。推测家蚕越年蚕种在初春中间感温中借助谷胱甘肽氧化还原代谢指标的变化抵抗温度变化,并为解除滞育做准备。这对于阐明蚕卵滞育解除和胚胎活化的生化机制具有一定理论意义。 Intermediate temperature sensing in the early spring is a technical step of double refrigeration for overwintering silkworm egg,and glutathione redox metabolism is closely related to diapause induction and termination of silkworm embryo.In order to understand physiological and biochemical changes in glutathione redox metabolism during the intermediate temperature sensing of silkworm egg,silkworm hybrid Jingsong×Haoyue(JS×HY)was taken as an experimental material to detect and compare its glutathione content and activity of enzyme related to glutathione redox cycle during the intermediate temperature sensing in the early spring.It was found that changes in activity of GSH(reduced glutathione)related enzyme has a certain effect on the resistance of silkworm egg to temperature injury,and diapause termination of silkworm egg needs high GR(glutathione reductase)activity.Besides,significant decrease of GR activity is one of the main reasons for the significant decrease of GSH/GSSG(oxidized glutathione)ratio.It is speculated that overwintering silkworm egg could resist the damage caused by temperature change during intermediate temperature sensing in the early spring and prepare for diapause termination by means of changing index of glutathione redox metabolism.It provides theoretical basis to understand biochemical mechanism of diapause termination of silkworm egg as wel as embryo activation.
