摘要
本研究将栉孔扇贝(Chlamys farreri)分别暴露于0、0.001、0.01和0.1μg/L的2,3,7,8-四氯二苯并-对-二噁英(TCDD)中,分别于0、1、3、6、10、15d取消化盲囊样品,初步探究了栉孔扇贝在TCDD胁迫下的解毒代谢过程。结果表明:TCDD对栉孔扇贝AhR信号通路和解毒代谢关键基因表达均具有显著影响,而对照组无明显变化。各染毒组AhR信号通路和I相、II相、III相代谢关键基因表达在15d内呈峰值变化,于10d达到最大值,其中HSP90、XAP2和ABCB1基因表达量呈现明显剂量效应,GST-microsomal、SULT1B1、UGT和ABCC1、ABCG2基因表达量均为:中浓度组>高浓度组>低浓度组,而且AhR、ABCC1、ABCG2基因表达分别与ARNT、GST-microsomal、UGT和SULT1B1基因表达变化趋势类似。IBR分析表明,解毒代谢关键基因表达在不同时间对TCDD染毒浓度表现出不同的响应水平,ARNT、ABCG2基因表达在各个染毒时间点均具有较高响应程度。由此可见,栉孔扇贝在TCDD胁迫下由AhR信号介导,通过I相、II相、III相连续的代谢完成对TCDD的解毒代谢过程,ARNT、ABCG2基因表达可作为栉孔扇贝在TCDD胁迫下的生物标志物。
To preliminarily investigate the detoxification mechanism of scallop Chlamys farreri exposed to 2,3,7,8-tetrachlorodibenzo-p-dioxin(TCDD),C.farreri was exposed to TCDD at 0,0.001,0.01 and 0.1μg/L and digestive gland was sampled on 0,1,3,6,10 and 15 d after initiation of the exposure.Results demonstrated that the TCDD exposure had a significant effect on the mRNA abundance of AhR signaling pathway and detoxification-related genes while no significant change was found in control groups.All the genes tested showed a peak change during the entire experiment and the highest levels occurred at 10 d.The expression levels of HSP90,XAP2 and ABCB1 genes showed significant dose-dependent manners,and the highest expression levels of GST-microsomal,SULT1 B1,UGT and ABCC1,ABCG2 genes were found in 0.01 g/L treatments while the lowest expression levels of them were found in 0.001 g/L treatments.The mRNA abundance trends of AhR,ABCC1 and ABCG2 genes were similar to those of ARNT,GST-microsomal and UGT,SULT1 B1 genes,respectively.The IBRv2 results revealed that the expression levels of ARNT and ABCG2 genes owned higher levels during the entire experiment.Therefore,we suggested that TCDD detoxification of C.farreri was mediated by AhR signaling pathway,and involved continuous metabolism of phase I,phase II and phase III.In addition,the expression levels of ARNT and ABCG2 genes could be used as the biomarkers of C.farreri under TCDD exposure.
