摘要
目的考察CYP2D6^(*)10等位基因在肝癌患者中的分布,探讨^(*)10等位基因与肝癌的易感性。方法收集50例肝细胞肝癌(HCC组)和50例肝血管瘤患者(对照组)的肝组织和血液标本,进行组织病理学分级,采用聚合酶链反应进行DNA扩增,采用质谱测序进行CYP2D6^(*)10基因分型。结果与对照组相比,HCC组CYP2D6^(*)10等位基因分布存在显著性差异,突变纯合子TT频率显著降低(41.4%vs.64.3%,P<0.05),杂合子CT频率升高(24.1%vs.7.1%,P<0.05)。50例HCC肝标本中,24例为肝纤维化,26例为肝硬化,肝硬化和纤维化组与对照组相比,TT频率显著降低。结论肝癌病人CYP2D6^(*)10等位基因频率显著降低,^(*)10突变可能与肝癌易感性具有相关性。
Objective To investigate the frequency of CYP2D6^(*)10 allele in the patients with hepatocellular carcinoma and the susceptibility of gene mutation to liver cancer.Methods Liver tissue and blood samples were collected from 50 patients with hepatocellular carcinoma(HCC group)and 50 patients with hemangioma(control group).Histopathological grading was performed.The DNA was amplified by Polymerase Chain Reaction and the CYP2D6^(*)10 genotype was determined by Mass spectrum.Results Compared with the control group,the frequency distribution of CYP2D6^(*)10(100 C>T)allele in hepatocellular carcinoma group was significantly difference.The frequency of homozygous TT mutation significantly reduced in hepatocellular carcinoma patients(41.4%vs.64.3%,P<0.05).The frequency of heterozygous CT increased in patients than in controls(24.1%vs.7.1%,P<0.05).Among the 50 HCC liver specimens,24 cases showed liver fibrosis and 26 cases showed cirrhosis.TT frequency was also significantly reduced in the cirrhosis and fibrosis groups compared with the control group.Conclusion The mutation frequency of CYP2D6^(*)10 decreased significantly in patients with liver cancer.The^(*)10 mutation may be related to the incidence of liver cancer.
