摘要
目的探究miR-370-3p对结肠癌细胞增殖和迁移的影响及可能机制。方法转染结肠癌SW620细胞,分为空白对照组,阴性对照组和miR-370-3p模拟物组,实时荧光定量PCR检测miR-370-3p和ANO1 mRNA表达,Western blot检测ANO1蛋白表达,通过CCK-8法检测miR-370-3p对细胞增殖活性的影响,Transwell法检测miR-370-3p对细胞迁移的影响,生物信息学网站预测ANO1是否为miR-370-3p的潜在靶基因,双荧光素酶报告基因实验进行验证。结果转染miR-370-3p模拟物后,SW620细胞ANO1 mRNA和蛋白表达降低,SW620细胞的增殖和迁移被抑制,ANO1是miR-370-3p的靶基因。结论miR-370-3p通过靶向调控ANO1抑制结肠癌细胞增殖和迁移。
Objective To explore the effect and possible mechanism of miR-370-3p on the proliferation and migration of colon cancer cells.Methods The SW620 cells were divided into blank control group,negative control group and miR-370-3p mimic group.The expressions of miR-370-3p and ANO1 mRNA were detected by qRT-PCR,and the expression of ANO1 was detected by Western blot.CCK-8 assay was used to detect the effect of miR-370-3p on cell proliferation activity,and Transwell assay was used to detect the effect of miR-370-3p on cell migration.Bioinformatics website predicted whether ANO1 was a potential target gene of miR-370-3p,and dual luciferase reporter gene assay was used to verify the results.Results After transfection with miR-370-3p mimic,the mRNA and protein expressions of ANO1 in SW620 cells were decreased,and the proliferation and migration of SW620 cells were inhibited in vitro,ANO1 was the target gene of miR-370-3p.Conclusion miR-370-3p inhibited the proliferation and migration of colon cancer cells by targeting ANO1.
