摘要
用液闪计数测定经组织培养的6株人肿瘤(MGC 80-3、SGC-7901、BEL-7402、Os-732、Detroit-6和HeLa)细胞及1株正常成纤维细胞对^(3)H-HPD的摄取和存留。MGC 80-3和成纤维细胞的摄取量,在测定的24h内,随温育时间延长而增加;温育30 min换液,再培养30 min,血卟啉衍生物含量就明显减少,MGC 80-3和成纤维细胞分别只含原摄入量的32.01%和40.75%;但在以后的24h内,维持在类似的水平上。7株细胞温育30min摄入^(3)H-HPD量,按多少顺序依次为成纤维细胞、HeLa、BEL-7402、Detroit-6、SGC-7901、MGC 80-3及Os-732;但是,培养24h后,由于各株细胞排出量不同,^(3)H-HPD存留率大小顺序为Os-732、Detroit-6、BEL-7402、HeLa、SGC-7901,MGC 80-3及成纤维细胞。环境因素对细胞摄取^(3)H-HPD起重要作用,提高pH值、加入血清或降低温度都使摄入减少。本文结果提示,恶性组织比正常组织血卟啉衍生物含量高,不是因为恶性细胞摄入多,而是由于存留时间长,与体内实验结果一致,可供HPD光敏治疗恶性肿瘤作参考。
The uptake and retention of^(3)H-HPD in.human malignant cells(cell line MGC 80-3,SGC-7901,BEL-7402,Os-732,Detroit-6 and HeLa)and normal fibroblasts were studied by liquid scintillation counting method.The order of cell lines which took up^(3)H-HPD after 30 min.incubation were fibroblasts,HeLa,BEL-7402,Detroit-6,SGC-7901,MGC 80-3 and Os-732 respectively according to the amount of^(3)H-HPD in cells.Due to the variety in the amount of excretion,however,those cells which retained^(3)H-HPD after 24 hours cultivation were Os-732,Detroit-6,BEL-7402,HeLa,SGC-7901,MGC 80-3 and fibroblasts respectively in respect to the^(3)H-HPD level in cells.The uptake was reduced by raising extracel-lular pH and lowering temperature as well as adding higher concentration of serum.Our results showed that the^(3)H-HPD content in malignant ceils was higher than those in normal ones and it was because that malignant cells retained^(3)H-HPD longer other than took up more.
