摘要
目的:探讨LncRNA-00311与HOXC10信号通路的关系及其对口腔鳞癌侵袭转移的影响。方法:以HaCaT细胞和4种人口腔鳞癌细胞株(HN4、Cal27、HN30、HN6)为细胞模型,LncRNA-00311过表达病毒感染肿瘤细胞后,用RT-PCR、蛋白质印迹法等检查相关基因及蛋白表达情况,同时采用划痕实验检测肿瘤细胞侵袭迁移能力的变化。结果:LncRNA-00311过表达的HN6细胞的迁移能力显著降低,同时HOXC10的基因及蛋白表达显著下降,E-cadherin蛋白表达显著上调,N-cadherin和Vinmentin显著下调。结论:LncRNA-00311可能通过降低HOXC10表达,抑制HOXC10信号通路,进而抑制EMT,从而降低口腔鳞癌侵袭和转移。
Objective:To investigate the relationship between LncRNA-00311 and HOXC10 signaling pathway and its effect on invasion and metastasis of oral squamous cell carcinoma(OSCC).Methods:HaCaT cells and four human oral squamous cell lines(HN4,Cal27,HN30,and HN6)were used as cell models.After LncRNA-00311 overexpressed virus infected tumor cells,the expression of related genes and proteins were examined by RT-PCR and Western Blot,and the invasion and migration ability of tumor cells were detected by Wound Healing.Results:LncRNA-00311 over-expressed HN6 cells showed significantly decreased migration ability,while HOXC10 gene and protein expression were significantly decreased,E-cadherin protein expression was significantly up-regulated,and N-cadherin and Vinmentin were significantly down-regulated.Conclusion:LncRNA-00311 may reduce the expression of HOXC10 and inhibit the HOXC10 signaling pathway,thereby inhibiting EMT,invasion and metastasis of OSCC.
