摘要
目的分析急性髓系白血病(AML)来源骨髓间充质干细胞(AML-MSC)和健康人骨髓间充质干细胞(HD-MSC)基因表达谱和细胞因子表达的差异,并探讨AML-MSC在肿瘤微环境中的作用。方法从AML患者和健康人中获取MSC,分为AML-MSC组(4例)和HD-MSC组(4例)。采用流式细胞仪检测两组细胞表面抗原表达水平。对两组细胞进行成骨分化、成脂分化、转录组测序。采用ELISA法检测MSC上清液中细胞因子水平。将AML-MSC与白血病细胞株HL-60共培养后检测细胞凋亡情况。结果AML-MSC组中的细胞形态学、成骨分化及成脂分化情况、表面抗原表达水平与HD-MSC组表现出相似特征。AML-MSC组中转化生长因子(TGF)-β_(1)、趋化因子8(CXCL8)、血管内皮细胞生长因子(VEGF)水平均高于HD-MSC组(P<0.05)。共培养组活细胞比例高于不共培养组(P<0.05)。结论AML-MSC表现出基因表达谱和细胞因子表达的多方面改变,这将有助于了解MSC在促进AML发展中的作用,为寻找AML治疗新靶点提供理论依据。
Objective To analyze the gene expression profiles and cytokine expression differences between bone marrow mesenchymal stem cells derived from acute myeloid leukemia(AML-MSC)and healthy human bone marrow mesenchymal stem cells(HD-MSC),and to explore the role of AML-MSC in the tumor microenvironment.Methods MSC from patients with AML and healthy people were divided into AML-MSC group(4 cases)and HD-MSC group(4 cases).The expression of cell surface antigen was detected by flow cytometry.Osteogenic differentiation,adipogenic differentiation and transcriptome sequencing were performed in the two groups.The cytokine levels in MSC supernatant were detected by ELISA.The cell apoptosis of AML-MSC cocultured with HL-60 was detected.Results The cell morphology,osteogenic differentiation,adipogenic differentiation and surface antigen expression of AML-MSC were similar to those of HD-MSC group.Transforming growth factor(TGF)-β_(1),chemokine 8(CXCL8),vascular endothelial growth factor(VEGF)levels in AML-MSC group were higher than those in HD-MSC group(P<0.05).The proportion of viable cells in co-culture group was higher than that in non-co-culture group(P<0.05).Conclusion AML-MSC show multiple changes in gene expression profile and cytokines expression,which will help to understand the role of MSC in contributing to the development of AML and provide theoretical basis for finding new therapeutic targets for AML.
