蜈蚣败毒饮对银屑病模型鼠细胞标志物影响相关性研究 被引量:3

摘要 目的对蜈蚣败毒饮干预后的血清学中T淋巴细胞、树突状细胞、NK细胞、巨噬细胞、内皮细胞的细胞标志物进行检测,同时对其与银屑病模型鼠皮损指数的相关性进行分析。方法采用咪喹莫特诱导的银屑病大鼠为研究对象,将其随机分为空白对照组、蜈蚣败毒饮低剂量组、蜈蚣败毒饮中剂量组、蜈蚣败毒饮高剂量组、甲氨蝶呤西药对照组,经灌胃治疗后,采用改良的PASI评分法对银屑病模型鼠的皮损指数进行评价,采用流式细胞法对各免疫细胞的相关标志物CD3、CD4、CD8、CD11c、CD56、CD163、CD31进行检测,同时分析蜈蚣败毒饮各剂量组灌胃前后皮损指数变化量(△MPASI)与各细胞标志物变化量之间的相关性,从而分析蜈蚣败毒饮的细胞作用靶点。结果灌胃后各药物组大鼠的MPASI分值均有不同程度的下降(P<0.05或P<0.01),以甲氨蝶呤西药对照组和蜈蚣败毒饮高剂量组的效果最为显著(P<0.01),蜈蚣败毒饮中、高剂量组能够显著下调T淋巴细胞的标志物CD3和内皮细胞的标志物CD31(P<0.01),并显著上调T淋巴细胞的标志物CD4/CD8的比例(P<0.01),同时,蜈蚣败毒饮各剂量组干预后的皮损指数变化量(△MPASI)与T淋巴细胞标记物的变化量(△CD3和△CD4/CD8)及内皮细胞标记物的变化量(△CD31)具有正相关性(P<0.05)。结论蜈蚣败毒饮可以有效改善银屑病模型鼠的皮损状态,其机制可能与干预T淋巴细胞和内皮细胞的免疫途径有关。 Objective The cell markers of T lymphocytes,dendritic cells,NK cells,macrophages and endothelial cells in serum were detected after the intervention of Wugong Baidu Decoction(蜈蚣败毒饮),at the same time,the correlation between skin lesion index of psoriasis model mice and it was analyzed.Methods The psoriasis mice induced by imiquimod were used as the research object,they were randomly divided into blank control group,Wugong Baidu Decoction low,medium and high dose groups,and methotrexate Western medicine control group,after intragastric administration,the modified PASI score was used to evaluate the skin lesion index of psoriasis model mice,immune cell markers of CD3,CD4,CD8,CD11c,CD56,CD163 and CD31 were detected by flow cytometry,meanwhile,the correlation between the changes of skin lesion index(△MPASI)and the changes of cell markers before and after gavage in Wugong Baidu Decoction groups was analyzed,so as to analyze the cell target of Wugong Baidu Decoction.Results The MPASI scores of mice in each drug group decreased in different degrees after intragastric administration(P<0.05 or P<0.01),methotrexate western medicine control group and Wugong Baidu Decoction high dose group had the most significant effect(P<0.01),the medium and high dose of Wugong Baidu Decoction could significantly down regulate CD3 of T lymphocyte and CD31 of endothelial cell(P<0.01),and,the ratio of CD4/CD8 of T lymphocytes was significantly increased in both groups(P<0.01),at the same time,there were positive correlation between the changes of skin lesion index(△MPASI)and the changes of T lymphocyte markers(△CD3 and△CD4/CD8)or endothelial cell markers(△CD31)in Wugong Baidu Decoction groups after intervention(P<0.05).Conclusion Wugong Baidu Decoction can effectively improve the skin lesions of psoriasis model mice,the mechanism may be related to the immune pathway of T lymphocytes and endothelial cells.
