摘要
目的考察胆囊切除对小鼠肝细胞膜转运体胆盐输出泵(Bsep)表达及肝脏组织和血清中各胆酸盐含量的影响。方法制备胆囊切除小鼠模型,于术后第1、3、6周眼球采血、采集肝脏;通过液相色谱-质谱联用仪检测肝脏和血清中各胆酸盐浓度;采用Western blot技术和RT-qPCR技术检测肝细胞膜转运体Bsep的变化。结果胆囊切除小鼠术后体重等生命体征较假手术组无显著差异。术后3周,其肝细胞膜转运体Bsep表达较对照组显著降低(P<0.01),且肝脏中次级胆酸盐β-MCA、TCDCA、TDCA、TUDCA、THDCA质量分数较假手术组均显著升高,血清中次级胆酸盐β-MCA、CA、TCA、TCDCA、UDCA质量分数较假手术组均显著降低。结论胆囊切除可使小鼠肝脏中介导胆酸盐外排的转运体Bsep的表达降低,且其胆酸盐含量及成分比例发生了不同程度的改变,可能与胆囊切除术后患者再发结石的形成有关。
Objective To investigate into the effects of cholecystectomy on the expression of hepatobiliary membrane transporter Bsep and bile salts concentration in the liver and serum of mouse.Methods The cholecystectomy model was applied to the experiment,sampling blood was collected from eyes and hepastic tissue one,three,six weeks after operation;the concentrations of bile acid in liver and serum were determined by LC-MS/MS,the protein mass of Bsep was analyzed by Western blot and RT-qPCR.Results In contrast to the sham-operation,the vital signs did not show prominent changes,the expression of hepatobiliary membrane transporter Bsep was reduced(P<0.01)and the bile salts concentration in liver,includingβ-MCA,TCDCA,TDCA,TUDCA and THDCA,was increased and in serum,includingβ-MCA,CA,TCA,TCDCA and UD-CA,was reduced and part of them were changed observably(P<0.05)in the third week.Conclusions The cholecystectomy may cause the expression of the transporter Bsep to the effect that lead bile salts would be outwardly reduced.Meanwhile,the bile salts concentration and partition ratio in liver and serum of mouse may change at different levels,which could be associated with a postoperative bile duct calculi recurrence.
