摘要
以CMC-Na为唯一碳源,采用平板分离法从河套酒业酒糟中分离筛选出1株产低温酸性纤维素酶菌株C-11,16S rDNA基因序列与枯草芽孢杆菌的同源性较高。菌株C-11所产纤维素酶的最适反应温度为30℃,在35℃保持97%的活性,30℃保温24 h可保持91%的酶活力,保温60 h保持67%的活性,热稳定性好;该酶在pH 5.0时酶活最高,在pH 4.0和pH 5.0下保存60 h可保持38%和40%的活性。菌株C-11所产纤维素酶为低温酸性纤维素酶,可以耐受酒糟环境,降低酒糟纤维素含量。
A bacterium which produces cold-adapted acidity cellulase was isolated from Distillers’ grains on a selective medium that contained CMC-Na as the only source of carbon.The sequence of 16 S rDNA of strain C-11 showed 99% homology to Bacillus subtilis. The cellulase exhibited maximum activity at 30℃,retained 97% of its maximum activity at 35℃,The cellulase retained 91% and 67% of its activity for 24 and 60 h at 30℃,and the thermal stability is good. The maximal activity was observed at pH 5.0,Saving 60 h at pH 4.0 and 5.0 can maintain 38% and 40% of acitivity.The rsesults show that the cellulase produced by strain C-11 is cold-adapted acidity cellulase, which can tolerate the Distillers’ grains and reduce the cellulose of the Distillers’ grains.
