摘要
目的对2015—2019年河南省急性弛缓性麻痹(acute flaccid paralysis,AFP)病例中,在转人类脊髓灰质炎病毒受体小鼠肺细胞(mouse cell line expressing the gene for human cellular receptor for poliovirus,L20B)增殖的非脊髓灰质炎肠道病毒基因定型和序列特征分析。方法利用VP4-VP2编码区引物EVP4和OL68-1进行肠道病毒鉴定,均为柯萨奇A组病毒(coxsackievirus A,CV-A),分别为CV-A4、CV-A8和CV-A10。在利用CV-A4、CV-A8和CV-A10 VP1区特异性引物进行VP1区序列扩增和亲缘进化树构建。结果9例L20B阳性分离物中,1例为CV-A4,1例为CV-A8,7例为CV-A10。通过亲缘进化树构建,CV-A4属于D基因型,CV-A8属于B基因型,CV-A10属于E基因型。结论河南省L20B细胞增殖的非脊髓灰质炎肠道病毒为CV-A4、CV-A8和CV-A10,通过这些病毒基因特征分析对手足口病和AFP预防控制具有重要意义。
Objective To isolate non-polio enteroviruses from patients with acute flaccid paralysis(AFP)from 2015 to 2019 in Henan Province,and to identify and genetically characterize the isolates proliferated on L20 B cells(mouse cell line expressing the gene for human cellular receptor for poliovirus).Methods The primers used for EVP4 and OL68-1 which were based on VP4-VP2 gene located in the coding region,were designed to isolate from coxsackievirus A(CV-A),including CV-A4,CV-A8 and CV-A10.Full-length VP1 genes of the three isolates were amplified by RT-PCR.The VP1 gene homology was analyzed by Megalign software.Results In the experiment,9 isolates were identified,including one case of CV-A4,one case of CV-A8,and 7 cases of CV-A10.Phylogenetic tree analysis indicated that CV-A4 belonged to D genotype,CV-A8 belonged to B genotype,and CV-A10 belonged to E genotype.Conclusion The CV-A viruses multiplied on L20 B cells,and caused cytopathic effect.The genetic characteristics showed that CV-A4,CV-A8 and CV-A10 were predominant CV-A viruses identified in Henan Province,these viruses are of great significance in the prevention and control of hand,foot and mouth disease and AFP.
