矽尘对大鼠脂质过氧化及肺纤维化的影响 被引量:2

The effects of silica dust on lipid peroxidation and pulmonary fibrosis in rats

摘要 目的探讨矽尘对大鼠脂质过氧化及肺纤维化的影响。方法40只大鼠随机分为对照组和30、60、120 d实验组,每组10只。实验组采用一次性染尘法,经气管向大鼠肺内注入1 ml二氧化硅(SiO2,100 mg/ml)悬浊液,分别于染尘后30、60和120 d处死,对照组不染尘(注入生理盐水),饲养120 d后处死,其他操作同实验组。采集大鼠血清并摘取双肺。左肺用于电子显微镜观察;右肺行苏木素-伊红(HE)和马松(Masson)染色,观察肺组织病理变化。同步分光光度法测血清中丙二醛(MDA)、超氧化物歧化酶(SOD)、一氧化氮(NO)、谷胱甘肽(GSH-Px)的含量。结果与对照组相比,3个实验组胶原纤维增生明显,染尘时间越长,增生越明显。与对照组相比,各染尘组大鼠血清MDA和NO含量均升高(P<0.05),30 d组尤为明显。与对照组相比,实验组SOD活力降低,GSH-Px活力升高,差异均有统计学意义(P<0.05)。结论在一次性大剂量矽尘暴露所致的矽肺纤维化进程中,30 d时脂质过氧化严重,但纤维化程度较轻;120 d时脂质过氧化较轻,但纤维化程度较重。 Objective To investigate the effects of silica dust on lipid peroxidation and pulmonary fibrosis in rats.Method Forty rats were randomly divided into a control group and 30 d,60 d,120 d experimental groups with 10 rats in each group.The rats of experimental groups were injected with 1 ml SiO2(100 mg/ml)suspension through the trachea only once,thenwere executed at 30 days,60 days,120 days after dust-staining respectively.The control group rats were not stained with silica dust(injected with physiology saline),and killed after 120 days.The serum and lung were taken and the left lung was observed by electron microscope.The right lung tissue was taken to stain with HE and Masson method,then was the pathological changes was observed by microscope.At the same time,the contents of MDA,SOD,NO and GSH-Px in serum were measured by spectrophotometry.Results Compared with the control group,manted protiferation of collagen fibers were observed in the three experimental groups.The longer time after dust exposure,the more hyperplasia were observed.Compared with the control group,the levels of MDA,and NO in serum of rats in each dust-exposed group were increased(P<0.05).Compared with the control group,the activity of SOD decrease,while the activity of GSH-Px was increased(P<0.05).Conclusion In the process of silicosis fibrosis induced by one-time high-dose silica dust exposure,lipid peroxidation is serious in 30 d group,but fibrosis is mild;while the lipid peroxidation is mild,but fibrosis is serious in the 120 d group.
