IL-9在小鼠肺结核模型中对Treg的调控作用

摘要 目的研究白细胞介素(IL)-9在小鼠肺结核模型中对T调节细胞(Treg)的作用。方法建立结核菌感染小鼠模型,致敏的CD4+T细胞(Teff)与Treg按1∶1、1∶2、1∶4、1∶16进行培养,选取最适比例,设置对照组、Treg组、IL-9抗体组、生长转化因子β(TGF-β)抗体组、IL-10抗体组和IL-9组,应用流式细胞术检测培养细胞中Treg比例,ELISA法检测细胞培养上清液中IL-2、IL-4、IL-5、IL-9、IL-17、IL-10、TGF-β和干扰素-γ(IFN-γ)浓度,CCK-8法检测细胞增殖情况。结果Teff与Treg最适比例为1∶4时抑制效果最好。IL-9组Treg细胞占CD4+T细胞比例明显高于对照组、IL-9抗体组、IL-10抗体组和TGF-β抗体组,IL-9组对Treg抑制率分别高于IL-9抗体组和TGF-β抗体组,差异均有统计学意义(P<0.05)。与对照组比较,Treg组、TGF-β抗体组和IL-9组培养上清液中IFN-γ和IL-2的表达明显降低(P<0.05),与Treg组比较,IL-9加入抗体后IFN-γ和IL-2的表达明显增高(P<0.05)。IL-9组培养上清液中IFN-γ和IL-2浓度明显低于IL-10抗体组,IL-4、IL-5和IL-9的浓度明显高于对照组、IL-9抗体组、IL-10抗体组和TGF-β抗体组,差异均有统计学意义(P<0.05)。IL-9组培养上清液中IL-17的浓度明显低于对照组、IL-10抗体组和TGF-β抗体组,IL-10和TGF-β的浓度明显高于对照组、IL-9抗体组、IL-10抗体组和TGF-β抗体组,差异均有统计学意义(P<0.05)。结论IL-9在小鼠肺结核模型中具有促进Treg细胞增殖的作用,抑制Th1、Th17型细胞因子分泌IFN-γ、IL-2、IL-17,促使Th2型和Treg细胞相关因子IL-4、IL-5、IL-10和TGF-β分泌。 Objective To study the effect of interleukin(IL)-9 on T-regulatory cells(Treg)in a mouse model of pulmonary tuberculosis.Methods The mice model of tuberculosis infection was established.The sensitized CD4+T cells(Teff)and Treg culture were mixed in ratios of 1∶1,1∶2,1∶4 and 1∶16,and the optimal proportion was selected.The control group,Treg group,IL-9 antibody group,transforming growth factor-β(TGF-β)antibody group,IL-10 antibody group and IL-9group were set.Flow cytometry was used to detect the proportion of Treg in cultured cells;ELISA was used to detect the concentrations of IL-2,IL-4,IL-5,IL-9,IL-17,IL-10,TGF-βand interferon-γ(IFN-γ)in cell culture supernatant,and CCK-8 method was used to detect the cell proliferation.Results When the optimal ratio of Teff to Treg was 1∶4,the inhibition effect was the best.The proportion of Treg cells in CD4+T cells in IL-9 group was significantly higher than that in control group,IL-9 antibody group,IL-10 antibody group and TGF-βantibody group(P<0.05).The inhibition rate of Treg in IL-9 group was higher than those in IL-9 antibody group and TGF-βantibody group,respectively(P<0.05).Compared with the control group,the expressions of IFN-γand IL-2 in culture supernatant of Treg group,TGF-βantibody group and IL-9 group were significantly decreased(P<0.05).Compared with the Treg group,the expressions of IFN-γand IL-2 were significantly increased after the addition of antibody in IL-9 group(P<0.05).The concentrations of IFN-γand IL-2 in the culture supernatant of IL-9 group were significantly lower than those of IL-10 antibody group(P<0.05).The concentrations of IL-4,IL-5 and IL-9 in the supernatant of IL-9 group were significantly higher than those in the control group,IL-9antibody group,IL-10 antibody group and TGF-βantibody group(P<0.05).The concentration of IL-17 in the supernatant of IL-9 group was significantly lower than those of the control group,the concentration of IL-10 antibody group and TGF-βantibody group(P<0.05);the concentration of IL-10 and
