摘要
本文报道了在中国内地首次发现的一例Hb G-Chinese(α230(B11)Glu→Glnβ2),并应用高压液相层析和微量双偶合法等技术对其一级结构进行了分析。
A slow-moving hemoglobin was demonstrated by electrophoresis in the blood of aChinese girl born in Chengdu,Sichuan Province.No clinical manifestations were observ-ed.The variant was present in 12.1% of the total hemoglobin.The propositus’ globinwas isolated and digested with TPCK-trypsin.High pressure liquid chromatographicseparation of the tryptie peptides revealed a slower eluting αT-4 peptide.The amino acidanalysis of this zone was normal.The peak was sequenced by the DABITC/PITC doublecoupling method.The result indicated that glutamic acid residue at position 30 of thechain was subsituted by a glutaminyl residue.The abnormal hemoglobin was thus iden-tified as Hemoglobin G-Chinese(α30 Glu→Gln β).This is the first case of Hb G-Chinese found in the mainland of China.
