乳腺癌类器官研究进展及中医药应用展望

摘要 乳腺癌是一种高度异质性的恶性肿瘤,其包括多种不同的亚型,这些亚型在遗传、病理、临床治疗及预后上各不相同。传统的乳腺癌细胞系研究模型不能完全符合临床肿瘤发病的实际病理过程及其肿瘤异质性的特征。乳腺癌类器官(Organoids)是一种基于3D细胞培养(Three-dimensional cells culture)系统建立的模型,在保留患者肿瘤的分子表型和基因型变化的同时,再现原发肿瘤的异质性,因此被应用于肿瘤发病机制和抗肿瘤药物筛选的研究中。目前,乳腺癌的中医药研究大多以细胞系和人源性肿瘤异种移植为主,类器官的相关研究则相对缺乏。该综述简要介绍乳腺癌类器官培养体系的发展,以及其在生命科学研究中的应用,以期为中医药乳腺癌相关研究提供思路。 Breast cancer is a highly heterogeneous malignancy,including a variety of different subtypes,which differ in genetics,pathology,clinical treatment and prognosis.The traditional research model of breast cancer cell lines cannot fully conform to the actual pathological process of clinical tumor pathogenesis and the characteristics of tumor heterogeneity.Breast cancer organoid is a model based on 3D cell culture system,which can reproduce the heterogeneity of the primary tumor while retaining the molecular phenotype and genotype changes of the patient′s tumor.Therefore,it has been applied in the study of tumor pathogenesis and anti-tumor drug screening.At present,the traditional Chinese medicine research on breast cancer is mainly focused on cell lines and patient-derived tumor xenografts,while the related research on organoids is relatively lacking.This review briefly introduces the development of breast cancer organoid culture system and its application in life science research,in order to provide ideas for breast cancer related research with traditional Chinese medicine.
