野木瓜总皂苷及皂苷单体对乳腺癌细胞增殖与迁移作用探讨

摘要 目的探究野木瓜总皂苷在乳腺癌细胞增殖迁移中的作用及其关键成分。方法选择人乳腺癌细胞MCF-7、人乳腺癌细胞MDA-MB-231、小鼠乳腺癌细胞4T1以及小鼠黑色素瘤细胞B16-F1,CCK-8实验检测不同浓度下野木瓜总皂苷及其单体对各肿瘤细胞增殖的影响;细胞划痕实验检测野木瓜单体YF-32对人乳腺癌细胞MCF-7迁移能力的影响。结果细胞增殖检测结果表明,与空白对照组相比,野木瓜总皂苷对以上各乳腺癌细胞增殖都有明显的抑制效果,并且其关键的有效成分是单体YF-32。细胞划痕实验结果表明,野木瓜总皂苷单体YF-32明显抑制了人乳腺癌细胞MCF-7的迁移。此外,野木瓜总皂苷对小鼠黑色素瘤细胞B16-F1的增殖也具有明显的抑制作用。结论野木瓜总皂苷对人乳腺癌细胞MCF-7、人乳腺癌细胞MDA-MB-231、小鼠乳腺癌细胞4T1的增殖都有很好的抑制效果,且其主要功能成分是单体YF-32。并且YF-32抑制了人乳腺癌细胞MCF-7的迁移。此外,野木瓜总皂苷对小鼠黑色素瘤细胞B16-F1增殖同样具有较好的抑制效果,说明野木瓜总皂苷很可能具有广谱抗癌作用。 Objective To investigate the effects and key components of the triterpenoid saponins from Stauntonia chinensis on proliferation and migration of breast cancer cells.Methods Human breast cancer cell MCF-7,human breast cancer cell MDA-MB-231,mouse breast cancer cell 4T1 and mouse melanoma cell B16-F1 were selected.The effects of triterpenoid saponins and monomers of Stauntonia chinensis on the proliferation of various tumor cells were detected by CCK-8.Cell scratch test was used to detect the effect of triterpenoid saponins monomer of Stauntonia chinensis the migration ability of tumor cells.Results Cell proliferation assay showed that compared with the blank control group,the triterpenoid saponins from Stauntonia chinensis had obvious inhibitory effect on the proliferation of breast cancer cells,and the key active component was monomer YF-32.The results of cell scratch test showed that the triterpenoid saponins from Stauntonia chinensis YF-32 significantly inhibited the migration of human breast cancer cell MCF-7.In addition,the triterpenoid saponins from Stauntonia chinensis also had significant inhibitory effect on the proliferation of mouse melanoma cells B16-F1.Conclusion The triterpenoid saponins from Stauntonia chinensis have good inhibitory effect on the proliferation of human breast cancer cell MCF-7,human breast cancer cell MDA-MB-231 and mouse breast cancer cell 4T1,and the main functional component of triterpenoid saponins from Stauntonia chinensis is YF-32.Moreover,YF-32 inhibited the migration of human breast cancer cells MCF-7.In addition,the triterpenoid saponins from Stauntonia chinensis also show good inhibitory effect on the proliferation of mouse melanoma cells B16-F1,suggesting that the triterpenoid saponins from Stauntonia chinensis may have a broad-spectrum anticancer effect.
