摘要
目的:探讨miR-216a在子宫肌瘤中表达及调控CAPN6基因对子宫肌瘤细胞增殖和凋亡的影响。方法:通过实时荧光定量聚合酶链反应(qRT-PCR)检测miR-216a在正常子宫肌层及子宫肌瘤组织和细胞中的表达。在子宫肌瘤细胞中过表达miR-216a,利用qRT-PCR检测CAPN6 mRNA的表达,Western blot法检测CAPN6蛋白的表达。利用Cell Counting Kit-8(CCK-8)实验检测过表达miR-216a后子宫肌瘤细胞增殖能力。流式细胞术检测过表达miR-216a后细胞凋亡情况。结果:miR-216a在子宫肌瘤组织中的表达低于子宫肌层组。过表达miR-216a后CAPN6基因mRNA及蛋白的表达量明显降低。过表达miR-216a抑制了子宫肌瘤细胞的增殖而促进了细胞凋亡。结论:miR-216a在子宫肌瘤中呈低表达,可在体外靶向抑制CAPN6基因,抑制子宫肌瘤细胞增殖能力,促进细胞凋亡,可能成为生物治疗子宫肌瘤的有效靶标分子。
Objective:To investigate the expression of miR-216 a in uterine fibroids and the effect of CAPN6 gene on proliferation and apoptosis of uterine fibroid cells.Methods:The expression of miR-216 a in normal myometrium,uterine fibroid tissues and cells was detected by real-time fluorescent quantitative polymerase chain reaction(qRT-PCR).miR-216 a was overexpressed in uterine fibroid cells,and the expression of CAPN6 mRNA was detected by qRT-PCR.The expression of CAPN6 protein was detected by Western blot.The proliferation of uterine fibroid cells after overexpression of miR-216 a was detected by Cell Counting Kit-8(CCK-8)assay.Flow cytometry was used to detect apoptosis after overexpression of miR-216 a.Results:The expression of miR-216 a in uterine fibroids was lower than that in the myometrium.The expression of CAPN6 mRNA and protein was significantly decreased after overexpression of miR-216 a.Overexpression of miR-216 a inhibited proliferation of uterine fibroid cells and promoted apoptosis.Conclusion:miR-216 a is lowly expressed in uterine fibroids,which can inhibit the CAPN6 gene in vitro,inhibit the proliferation of uterine fibroid cells and promote cell apoptosis,which may be an effective target molecule for biological treatment of uterine fibroids.
