摘要 目的探讨同型半胱氨酸(Homocysteine,Hcy)对血管内皮细胞衰老的影响。方法将人脐静脉内皮细胞(Human umbilical vein endothelial cells,HUVEC)用不同浓度Hcy处理24 h,通过细胞增殖活性检测、β-半乳糖甘酶染色、细胞周期检测对细胞进行分析。为进一步了解Hcy对细胞衰老的作用,使用白藜芦醇(Resveratrol,RSV)预处理后再进行Hcy干预。通过活性氧和端粒酶检测,探讨Hcy致血管内皮细胞衰老的作用。结果Hcy抑制细胞增殖活力和细胞分裂,增加细胞衰老程度,呈浓度依赖性。Hcy升高细胞内活性氧水平,降低端粒酶活性,RSV预处理后降低了活性氧水平,改善端粒酶活性和细胞衰老状态。结论Hcy通过活性氧增加加速内皮细胞衰老,这一过程与端粒酶失活有关。 Objective To explore the effect of Homocysteine(Hcy)on the aging of vascular endothelial cells.Method Human umbilical vein endothelial cells(HUVEC)were treated with different concentrations of Hcy for 24 h,and the cells were analyzed by cell proliferation activity detection,β-galactosidase staining,and cell cycle detection.In order to further understand the effect of Hcy on cell aging,resveratrol(RSV)pretreatment was used before Hcy intervention.Through the detection of reactive oxygen species and telomerase,the role of Hcy-induced vascular endothelial cell senescence was discussed.Results Hcy inhibited cell proliferation and cell division,and increased cell senescence in a concentration dependent manner.Hcy increased the level of reactive oxygen species and decreased the activity of telomerase.RSV pretreatment decreased the level of reactive oxygen species and improved telomerase activity and cell aging..Conclusion Hcy accelerates endothelial cell senescence by increasing reactive oxygen species,which is related to telomerase inactivation.
