职业性矽肺壹期患者血清生物标志物因子分析 被引量:1

Factor analysis on serum biomarkers in patients with stageⅠsilicosis

摘要 目的探讨血清趋化因子及氧化与抗氧化生物标志物在职业性矽肺(简称“矽肺”)中的作用。方法采用方便抽样方法,选择58例矽肺壹期患者为研究对象。采用酶联免疫吸附法检测其血清中核因子E2相关因子(Nrf2)、血红素氧合酶-1(HO-1)和8-异前列腺素F2α(8-iso-PGF2α)水平,采用化学比色法检测血清中脂质过氧化物(LPO)和总抗氧化能力(T-AOC),采用Luminex流式荧光技术检测血清中γ-干扰素诱导蛋白10(IP-10)、巨噬细胞炎症蛋白(MIP)-1α、MIP-1β和巨噬细胞衍生趋化因子(MDC)水平;对上述指标进行因子分析。结果9项生物标志物原始指标信息量提取率为58.5%~96.5%。提取到4个公因子分别为Nrf2抗氧化通路、辅助性T细胞(Th)1优势趋化、总的氧化/抗氧化平衡和Th2优势趋化,其方差贡献率分别为32.2%、19.1%、16.4%和11.8%,累计方差贡献率为79.5%。结论氧化/抗氧化失衡和Th选择性优势趋化均参与矽肺的发生发展;其中,Nrf2抗氧化通路的作用最为关键。 Objective To investigate the role of serum chemokines,and oxidative and antioxidant biomarkers in occupational silicosis(hereinafter referred to as silicosis).Methods A total of 58 patients with stageⅠsilicosis were selected as the research subjects using convenient sampling method.The serum levels of nuclear factor erythroid-2 related factor 2(Nrf2),heme oxygenase-1(HO-1)and 8-isoprstaglandin F2α(8-iso-PGF2α)were determined by enzyme-linked immunesorbent assay.The serum levels of lipid peroxide(LPO)and total antioxidant capacity(T-AOC)were determined by chemistry colorimetric method.Luminex flow fluorescence technology was used to detect the serum levels of interferon-γinducible protein-10(IP-10),macrophage inflammatory protein(MIP)-1α,MIP-1βand macrophage-derived chemokine(MDC).The above indicators were analyzed by factor analysis.Results The information extraction rate of the original indicators of the nine biomarkers was 58.5%-96.5%.Four common factors were extracted,including Nrf2 antioxidant signaling pathway,helper T cell(Th)1 dominant chemotaxis,the total oxidation/antioxidant balance,and Th2 dominant chemotaxis,whose variance contribution rates were 32.2%,19.1%,16.4%,and 11.8%,respectively,and the cumulative variance contribution rate was 79.5%.Conclusion Both the oxidant-antioxidant disturbance and the dominance chemotaxis are involved in the occurrence and development of silicosis,and the Nrf2 antioxidant signaling pathway plays the most critical role.
