摘要
[目的]观察糖尿病胃动力障碍大鼠胃窦平滑肌中蛋白激酶C(PKC)活性的变化情况.[方法]通过腹腔注射给予链脲佐菌素制作糖尿病大鼠模型,利用RM-6240四道生理信号记录仪观察大鼠胃窦环行平滑肌自发性收缩运动情况,将自发性收缩运动减弱的大鼠归为糖尿病胃动力障碍模型组.采用蛋白免疫印迹法观察PKC的活性变化.[结果]与正常对照组比较,糖尿病组大鼠胃窦平滑肌组织PKCα、PKCβⅠ蛋白膜浆比明显减小,且PKCα磷酸化情况亦明显降低(P<0.05),即PKC活性明显减弱.[结论]糖尿病大鼠发生胃动力障碍可能与胃窦平滑肌中PKC活性减弱有关系.
OBJECTIVE To observe the change of protein kinase C(PKC)activity in gastric antrum smooth muscle of diabetic gastric motility disorder rats.METHODS The diabetic rat model was made by intraperitoneal injection of streptozotocin.RM-6240 four-channel physiological signal recorder was used to observe the spontaneous contraction of smooth muscle in gastric antrum,and the rats with weakened spontaneous contraction were classified as diabetic gastric motility disorder model group.The activity of PKC was observed by western blot.RESULTS Compared with the normal control group,the membrane to cytoplasm ratio of PKCα and PKCβ Ⅰin the smooth muscle of gastric antrum of diabetic rats was significantly reduced and the phosphorylation of PKCα was also significantly decreased(P<0.05),in other words,the activity of PKC was significantly weakened.CONCLUSION Gastric motility disorder in diabetic rats may be related to the weakening of PKC activity in the smooth muscle of gastric antrum.
