摘要
[目的]探讨槭树科槭树属植物紫花槭中治疗败血症的天然有效化合物、潜在蛋白靶点及作用通路.[方法]运用网络药理学方法,采用多种天然产物分离技术建立紫花槭败血症疾病靶点数据集,构建紫花槭-化合物-疾病靶点-疾病蛋白通路网络图.[结果]共筛选出41种紫花槭中的活性成分,包括367个药物靶点和471个疾病作用靶点,其中涉及GO富集分析的生物过程、细胞组分及分子功能通路分别为2961、60及163条,涉及KEGG富集分析的通路为136条.[结论]紫花槭中含有作用于TNF通路中NF-κB、MAPK、P38、JUK等多种酶表达、抑制炎症反应并发挥治疗败血症作用的天然产物,可通过TNFMAPK通路减少炎症因子的释放起到抗败血症的作用.
OBJECTIVE To explore the natural effective compounds,potential protein targets and pathways of action of Acer pseudosieboldianum in the Aceraceae family in the treatment of septicemia.METHODS By using network pharmacology and various natural product separation techniques,a sepsis disease target dataset of Acer pseudosieboldianum was established,and the network diagram of the Acer pseudosieboldianum-compound-disease target-disease protein pathway was constructed.RESULTS A total of 41 active ingredients from Acer pseudosieboldianum were screened,including 367 drug targets and 471 disease action targets.Among them,there were 2961 biological processes,60 cell components,and 163 molecular functional pathways involved in GO enrichment analysis,and 136 pathways involved in KEGG enrichment analysis.CONCLUSION Acer pseudosieboldianum contains natural products that can act on the expressions of NF-κB,MAPK,P38,JUK and other enzymes in the TNF pathway,inhibit the inflammatory reaction and play a role in the treatment of sepsis,and play an anti-sepsis role by reducing the release of inflammatory factors through the TNF-MAPK pathway.
