艳山姜不同提取物对α-葡萄糖苷酶及DPP-4的抑制活性研究

摘要 目的:以α-葡萄糖苷酶和DPP-4作为降血糖研究靶点,筛选出艳山姜对其有抑制作用的活性部位。方法:对艳山姜的根、茎、叶三个部位采用系统溶剂提取法制备不同部位的提取物。测定不同提取物对α-葡萄糖苷酶和DPP-4体外酶活的影响。结果:大部分艳山姜提取部位对α-葡萄糖苷酶表现出抑制作用,半数抑制浓度(IC_(50))值在10μg/mL浓度水平左右,其中,根部的甲醇提取物显示出最大抑制效果,其IC_(50)值可达到0.813μg/mL。艳山姜对DPP-4也有一定抑制作用,大多数提取部位的IC_(50)值在100-500μg/mL之间,其中,茎的乙酸乙酯提取物显示出最大抑制效果,IC_(50)为157.201μg/mL。结论:艳山姜提取物对α-葡萄糖苷酶和DPP-4均有一定的抑制作用,这可能是其具有降糖作用的主要作用机制。可优先考虑从其根的甲醇提取物和茎的乙酸乙酯提取物中分离纯化出具有降血糖活性的先导化合物。 Objective:In this study,α-glucosidase and DPP-4 were selected as hypoglycemic targets to screen the active parts of Alpinia zerumbet(Pers.)Burtt.et Smith(AZ)with hypoglycemic effect.Method:The systematic solvent extraction method was used to extract the extracts from root,stem and leaf of AZ.The inhibitory effect of α-glucosidase and DPP-4 with AZ extracts were evaluated by extracellular test.Results:The majority of AZ extracts showed strong inhibition effect of α-glucosidase,and the IC_(50) values of most plant parts were around 10μg/mL concentration level.Among them,the methanol extract from the root presented the maximum inhibitory effect,and the half inhibitory concentration(IC_(50))was 0.813μg/mL.AZ extracts also has an inhibitory effect on DPP-4,and the IC_(50) value of most extracted parts was between100μg/mL to 500μg/mL.The ethyl acetate extract of the stem performed the largest inhibition effect,with 157.201μg/mL(IC_(50)).Conclusion:AZ has a beneficial impact on α-glucosidase and DPP-4 inhibition,which may be the mechanism of their hypoglycemic effect.In the further study,the methanol extract of AZ roots and the ethyl acetate extract of its stems should be given priority to separating and purifying the lead compounds with hypoglycemic activity.
