摘要
目的:通过网络药理学预测苎麻根治疗糖尿病出血潜在靶点的作用机制。方法:通过中药系统药理数据库和分析平台、GeneCards、TTD、OMIM等数据库获取苎麻根有效成分和已报道与糖尿病出血有关的靶标蛋白;利用Uniport数据库筛选获得相关基因靶点;将药物与疾病的靶点基因通过Venny软件获取映射基因;通过Metascape数据库进行GO和KEGG富集分析;最后利用Cytoscape 3.10.1软件将其可视化,构建“苎麻根有效成分-核心靶点蛋白-代谢通路”网络图,并分析互作关系。结果:通过筛选,获取苎麻根有效成分27个,相关潜在靶点177个,其中与调控糖尿病出血相关的靶点共23个;通过PPI网络并筛选的得到5个关键靶点,这些靶点主要涉及Pathway in cancer、Nitrogen metabolism等63条生物学通路。结论:苎麻根治疗糖尿病出血存在多成分、多靶点、多通路协同作用,为苎麻根进一步治疗糖尿病出血的研究提供理论基础和新思路。
Objective:To predict the potential target mechanism of ramie root in treating diabetic hemorrhage through network pharmacology.Methods:The active components of ramie root and the reported target proteins associated with diabetic bleeding were obtained by using TCM system pharmacological database and analysis platform,GeneCards,TTD,OMIM and other databases.Human related gene targets were screened using Uniport database.Target genes of drugs and diseases are obtained through Venny software mapping genes;The GO and KEGG enrichment analysis was conducted by Metascape database.Finally,Cytoscape 3.10.1 software was used to visualize it,and the network diagram of“effective components of ramie rootcore target protein-metabolic pathway”was constructed,and the interaction relationship was analyzed.Results:27 effective components and 177 potential targets of ramie root were obtained through screening,among which 23 targets were involved in the regulation of diabetic bleeding.Five key targets were obtained through PPI network screening,which mainly involved 63 biological pathways in cancer,Nitrogen metabolism and so on.Conclusion:There are synergistic effects of multi-component,multi-target and multi-pathway in the treatment of diabetic hemorrhage by Ramie root,which provides theoretical basis and new ideas for further research on the treatment of diabetic hemorrhage by Ramie root.
