摘要
目的研究橙皮苷对高脂饮食诱导肥胖小鼠脂肪组织脂质代谢的作用并探究其可能的分子机制。方法将40只雄性C57BL/6J小鼠随机分为正常(N)、高脂(HF)、低剂量橙皮苷干预[HP-L,200mg/(kg·d)]及高剂量橙皮苷干预[HP-H,400mg/(kg·d)]共4组,每组10只。灌胃干预12w后,比较各组小鼠体重变化,称量脂肪组织并计算肾周和附睾脂肪系数;酶法检测血清中TC、TG、HDL-C和LDL-C的含量;HE染色观察各组小鼠脂肪组织形态变化;Western blot检测附睾脂肪中PLIN1、HSL、ATGL和SREBF1蛋白的表达水平。结果与N组相比,HF组小鼠体重显著增加(P<0.05),血清TC、TG和LDL-C水平均明显上升(P<0.05),HDL-C水平下降(P<0.05),脂肪细胞体积变大,附睾脂肪中PLIN1、HSL和ATGL蛋白表达显著降低(P<0.05),SREBF1表达升高(P<0.05);与HFD组相比,HP-L组和HP-H组小鼠体重减轻(P>0.05,P<0.05),肾周、附睾脂肪重量和系数降低(P<0.05),且在HP-H组更加明显,血清TC、TG和LDL-C水平均显著下降(P<0.05),HP-L组HDL-C水平上升(P<0.05),HP-L组和HP-H组脂肪细胞体积明显缩小(P<0.05),附睾脂肪中PLIN1、HSL和ATGL蛋白表达显著升高(P<0.05),SREBF1表达降低(P<0.05)。结论橙皮苷能够通过调节脂质代谢相关蛋白表达,促进脂肪组织脂质分解,抑制脂质合成,从而改善肥胖小鼠脂质代谢异常,达到降脂减肥效果。
Objective To study the effect of hesperidin on lipid metabolism in adipose tissues from obese mice induced by high-fat diet and explore its possible molecular mechanism.Methods Forty male C57 BL/6 J mice were randomly divided into 4 groups(n=10):normal group(N),high-fat group(HF),low-dose hesperidin intervention group(HP-L,200 mg/(kg·d))and high-dose hesperidin intervention group(HP-H,400 mg/(kg·d)).After the intervention for 12 weeks,body weight and adipose tissue weight were compared and the perrenal and epididymal fat indexes were calculated.Enzymatic methods were used to detect serum TC,TG,HDL-C and LDL-C.HE staining was performed to observe the morphological changes of adipose tissue.Western blot was used to detect the protein expression of HSL,ATGL,PLIN1 and SREBF1 in epididymal fat tissues.Results Compared with the N group,the body weight of the HF group increased significantly(P<0.05),the levels of TC,TG,and LDL-C in serum increased significantly(P<0.05),the level of HDL-C decreased(P<0.05),and the volume of fat cells increased.The protein expression of PLIN1,HSL and ATGL in epididymal fat tissues was significantly reduced(P<0.05),and the expression of SREBF1 was increased(P<0.05).Compared with the HF group,mice in HP-L and HP-H groups lost weight(P>0.05,P<0.05),the weight and index of perrenal or epididymal fat tissues decreased(P<0.05),and more obvious changes were found in the HP-H group.The levels of TC,TG and LDL-C in the serum decreased significantly(P<0.05).The level of HDL-C increased in the HP-L group(P<0.05).The volume of adipocytes in the HP-L and HP-H groups was significantly reduced(P<0.05),the protein expression of PLIN1,HSL and ATGL in epididymal fat tissues was significantly increased(P<0.05),and the expression of SREBF1 was decreased(P<0.05).Conclusion Hesperidin can promote lipid decomposition in adipose tissues and inhibit lipid synthesis by regulating the expression of lipid metabolism-related proteins,and thereby improve the abnormal lipid metabolism of obese mice and lead to lipoly
