癌性血性胸水TIL细胞体外培养分析

摘要 目的研究并分析体外培养癌性血性胸水TIL细胞的具体状况。方法以我院2016年7月至2019年3月收治的癌性血性胸水40例患者为研究对象,根据IL-2、IL-15、IL-7及IL-21表达水平的高低分为实验组(表达水平低,20例),对照组(表达水平高,20例);均采用四甲基偶氮唑盐(MTT)方法检测TIL细胞体外细胞活性,使用酶联免疫(ELISA)检测方法检测白细胞介素-2(IL-2)、白细胞介素-5(IL-5)、白细胞介素-7(IL-7)及白细胞介素-21(IL-21)的水平,对比分析两组胸水TIL细胞体外情况。结果分别予以8小时、12小时的TIL细胞体外培养后,实验组TIL细胞体外增殖倍数均高于对照组,两组间存在差异性,统计学有意义(P<0.05);实验组TIL细胞体外抗肿瘤活性显著高于对照组,两组间存在差异性,统计学有意义(P<0.05)。结论对于癌性血性胸水患者而言,IL-2、IL-15、IL-7及IL-21表达水平的高低与TIL细胞体外增殖倍数和抗肿瘤活性有关,IL-2、IL-15、IL-7及IL-21表达水平低的患者体外增殖倍数多,抗肿瘤活性强。 Objective to study and analyze specific status of TIL cells in vitro culture of cancerous bloody pleural effusion.Methods treat 40 cases cancerous bloody pleural effusions patients admitted to our hospital from July 2016 to March 2019 as research objects,according to expression levels of IL-2,IL-15,IL-7 and IL-21 group,divide them into experiment group(low expression level,20 cases),control group(high expression level,20 cases).Detect TIL cell in vitro cell activity with tetramethyl azozole salt(MTT)method,levels of interleukin-2(IL-2),interleukin-5(IL-5),interleukin-7(IL-7)and interleukin-21(IL-21)with enzyme-linked immunoassay(ELISA).Compare in vitro pleural effusion TIL cells between two groups.Results after 8-hour and 12-hour TIL cell culture in vitro respectively,multiplication factor of TIL cells in experimental group was higher than control group,difference was statistically significant between two groups(P<0.05);anti-tumor activity in vitro was significantly higher than control group,difference was statistically significant between two groups(P<0.05).Conclusion expression levels of IL-2,IL-15,IL-7 and IL-21 are related to proliferation of TIL cells in vitro and anti-tumor activity of of cancerous bloody pleural effusion patients,those with low expression levels of IL-7 and IL-21 have multiple fold proliferation in vitro and strong antitumor activity.
