摘要
目的:优选三颗针有效成分最佳提取方法。方法:以盐酸小檗碱、盐酸药根碱、盐酸巴马汀的总量为综合考察指标,分别采用超声法、回流法、酶解法提取法。采用高效液相法测定盐酸小檗碱、盐酸药根碱、盐酸巴马汀含量,DIAMONSILC18色谱柱(250 mm×4.6 mm,5μm);流动相为乙腈-0.02 mol/L磷酸二氢钾溶液;检测波长265 nm;流速为1.0 mL/min;柱温为30℃。结果:回流法所提取3种生物碱总量最高。结论:回流法为提取三颗针生物碱有效成分最佳方法。
Objective:To optimize the best extraction method for the active ingredients of the Berberis.Methods:Use the total amount of berberine hydrochloride,jatrorrhizine hydrochloride,and palmatine hydrochloride as a comprehensive evaluation index.Compare the total amounts by using the ultrasonic extraction method,the reflux extraction method,or the enzymatic extraction method seperately.The method of High-performance liquid chromatography(HPLC)was used to measure the contents of berberine hydrochloride,jatrorrhizine hydrochloride and palmatine hydrochloride.A Diamonsil C18 column(250 mm×4.6 mm,5μm)was used for separation at 30℃with detection wavelength at 265 nm,the solvent(known as the mobile phase)was a mixture of acetonitrile and 0.02 mol/L potassium dihydrogen phosphate solution(the ratio of volume was X:Y),the flow rate was 1.0mL/min.Results:The total amount of the three alkaloids extracted by the reflux extraction method was the highest.Conclusions:Reflux extraction method is the best extraction method for the active ingredients of Berberis alkaloids.
