摘要
H9N2亚型禽流感病毒近年来在家禽中普遍发生,可造成蛋鸡产蛋量下降,肉鸡复合型呼吸道疾病,给养禽业造成了巨大的经济损失。本研究筛选保护谱广的H9N2亚型禽流感病毒流行株的HA、NA、M1基因,以共表达的方式构建重组杆状病毒。通过血凝活性检测、Western blot杂交、电镜观察等方法对蛋白表达及病毒样颗粒的组装进行鉴定。结果表明,构建的重组杆状病毒可同时表达HA、NA和M1蛋白,且表达的蛋白具有良好的血凝活性,电镜观察可见约100 nm的病毒样颗粒。将表达的蛋白制备油乳剂亚单位疫苗并免疫SPF鸡,免疫后诱导的HI抗体与全病毒灭活疫苗的HI抗体效价水平相当,且对异源毒株具有良好的交叉保护。
The H9N2 subtype of avian influenza virus(AIV)has spread widely in poultry populations in recent years,which causes egg production decrease,respiratory disease and great economic losses.In this study,HA,NA,and M1 genes of the AIV subtype H9N2 were screened and co-expressed to product the virus-like particles(VLPs)in the baculovirus expression system.Protein expression and assembly of virus-like particles were confirmed in hemagglutination,Western blot and electron microscopy.The results showed that the VLPs simultaneously expressed HA,NA and M1 proteins and had hemagglutination activity.The VLPs were about 100 nm as observed under electron microscope.Afterwards,SPF chickens were immunized with the VLP preparation for immunogenicity evaluation.The VLP preparation induced HI antibody response comparable to the current inactivated vaccine and cross protection against the heterologous strain.
