安石榴苷对脂多糖诱导的鸡腺胃上皮细胞损伤的保护作用机制 被引量:1

Protective effect of punicalagin on lipopolysaccharide-induced damage to chicken glandular gastric epithelial cells

摘要 鸡腺胃炎是鸡群一种临床常见的病症,主要表现为患病鸡生长阻滞,腺胃肿大、出血胃溃疡。安石榴苷(punicalagin,Pun)是石榴皮的主要活性成分,具有强抗氧化作用。为探究Pun对鸡腺胃上皮细胞损伤的保护及抗氧化作用机制,本研究通过体外培养鸡腺胃上皮细胞,用4 mg/L LPS处理细胞24 h,分别添加4,8,16μmol/L Pun处理鸡腺胃上皮细胞后24 h,检测细胞的损伤状态、氧化应激水平、胃黏膜保护因子、紧密连接蛋白的表达以及Nrf2信号通路的激活情况。结果显示,Pun提高了鸡腺胃上皮细胞活力,使其乳酸脱氢酶(LDH)的释放减少,活性氧(ROS)水平下降,超氧化物歧化酶(SOD)和谷胱甘肽过氧化物酶(GSH)水平上升,胃黏膜保护因子前列腺素E2(PGE2)、一氧化氮(NO)、三叶因子2(TFF2)、紧密连接蛋白(ZO-1)、咬合蛋白(occludin)、闭合蛋白(claudin-3),以及Nrf2信号通路相关蛋白Nrf2、醌氧化还原酶1(NQO1)和胞质接头蛋白(keap1)的表达水平显著上升;Nrf2抑制剂-ML385会减弱Pun的保护作用。综上,Pun可修复LPS诱导的鸡腺胃黏膜损伤,并通过抑制氧化应激实现对鸡腺胃上皮细胞的保护作用。 Chicken gastroenteritis is a clinically common disease in chickens,mainly manifested as stunted growth,swollen glandular stomach,bleeding ulcers,etc.The main active ingredient of pomegranate peel is punicalagin(Pun),which has extremely strong oxidation resistance.In order to explore the protective and antioxidant mechanism of Pun on the damage of chicken glandular epithelial cells,the damage status,oxidative stress level,gastric mucosal protective factor,expression of tight junction protein and activation of Nrf2signaling pathway were detected in chicken glandular epithelial cells cultured in vitro,the cells were treated with 4mg/L LPS for 24h,and then treated with 4,8,or 16μmol/L Pun for 24h.The damage status,oxidative stress level,expression of gastric mucosal protective factor,tight junction protein and activation of Nrf2signaling pathway were examined.The results showed that Pun increased the viability of chicken glandular gastric epithelial cells and decreased the release of lactate dehydrogenase(LDH);the level of reactive oxygen species(ROS)decreased and the levels of superoxide dismutase(SOD)and glutathione peroxidase(GSH)increased;the expression levels of the gastric mucosal protective factors prostaglandin E2(PGE2),nitric oxide(NO),triglyceride factor 2(TFF2),tight junction protein(ZO-1),occludin closure protein(claudin-3),and Nrf2signaling pathway-related protein Nrf2,quinone NADH dehydrogenase 1(NQO1)and cytoplasmic junction protein(Keap1)increased significantly.Nrf2inhibitor-ML385diminished the protective effect of Pun.In summary,Pun can repair LPS-induced damage of chicken glandular gastric mucosa and protect chicken glandular gastric epithelial cells by inhibiting oxidative stress.
