摘要
为了探究α-硫辛酸(α-LA)在镉(Cd)暴露的情况下对大鼠肾细胞MAPK通路的影响,以SD大鼠、大鼠肾小管上皮细胞系(NRK-52E)和大鼠原代肾小管上皮细胞(rPT)为试验模型,设置对照组、Cd暴露组、Cd+α-LA组以及α-LA组。采用透射电镜观察细胞超微结构的变化,用光学显微镜观察肾脏组织病理学的变化,通过Western blot检测MAPK信号通路相关蛋白的相对表达水平。结果显示,与对照组相比,Cd暴露组多数细胞出现细胞核固缩、变形、染色质边聚甚至溶解,细胞内出现大量空泡;HE染色可见细胞变性、细胞管型、管壁变薄甚至破裂的现象;ERK1/2、JNK1/2和p38的磷酸化水平显著或极显著升高。与Cd暴露组相比,Cd+α-LA组细胞核皱缩轻微,染色质分布相对均匀;肾小球毛细血管充血和肾小管变性明显改善;ERK1/2、JNK1/2和p38的磷酸化水平显著或极显著降低。α-LA组与对照组相比无明显差异。结果表明,Cd暴露可诱导大鼠肾细胞损伤并激活MAPK信号通路,而α-LA通过抑制MAPK信号通路的活化缓解Cd诱导的细胞损伤。
In order to investigate the effect ofα-lipoic acid(α-LA)on the MAPK pathway of rat kidney cells under cadmium(Cd)exposure.In this study,SD rats,rat renal tubular epithelial cell line(NRK-52E)and rat proximal tubular(rPT)cells were used as experimental models,and the control group,Cd exposure group,Cd+α-LA group,α-LA group were set up.The changes of cell ultrastructure were observed by transmission electron microscope,the pathological changes of kidney were observed by light microscope,and the expression levels of MAPK signaling pathway-related proteins were detected by Western blot method.The results showed that compared with the control group,most of the cells in the Cd-exposed group had pyknosis,deformation,chromatin edge aggregation and even dissolution,and a large number of vacuoles appeared in the cells;HE staining showed cell degeneration,wall thinning and even rupture and casts;the phosphorylation levels of ERK1/2,p38and JNK1/2were significantly or extremely significantly increased.Compared with the Cd-exposed group,the nucleus of the Cd+α-LA group was slightly shrunken,and the chromatin distribution was relatively uniform,in addition,glomerular capillary congestion and tubular degeneration were significantly improved,furthermore,the phosphorylation levels of ERK1/2,JNK1/2and p38were significantly or extremely significantly reduced.There was no significant difference between theα-LA group and the control group.The results showed that Cd exposure induce rat renal cell damage and activate MAPK signaling pathway,whileα-LA alleviate Cd-induced cell damage by inhibiting the activation of MAPK signaling pathway.
