摘要
旨在探究中国荷斯坦奶牛磷酸二酯酶10A(phosphodiesterase 10A,PDE10A)基因的多态性及其与奶牛乳品质性状的相关关系。对104头中国荷斯坦奶牛基因组DNA,采用PCR扩增技术获得PDE10A基因的目的片段,通过直接测序法筛选相关多态性位点,并将其与乳品质性状进行了关联分析。同时,应用Haploview 4.2软件对相关SNPs进行连锁不平衡分析获得单倍型,解析PDE10A基因单倍型与奶牛乳品质性状的关联性。研究结果发现,PDE10A基因内含子12、13中共检测出3个SNPs位点,其中I12-3228 A>G、I13-13 A>G位点与中国荷斯坦奶牛产奶量、乳脂肪、乳蛋白、乳糖、体细胞数、干物质含量及矫正奶量均呈显著相关(P<0.05)。H1H3单倍型的中国荷斯坦奶牛具有更高的产奶量以及较高的乳脂肪、乳蛋白及乳糖含量。本研究结果表明,PDE10A基因可作为影响中国荷斯坦奶牛产奶性状的候选基因用于动物育种的标记辅助选择。
The aim of this experiment was to investigate the polymorphism of phosphodiesterase 10A(PDE10A)gene and its association with milk quality traits in Chinese Holstein cows.Genomic DNA from 104Chinese Holstein cows was used to obtain the target fragment of PDE10Agene by PCR amplification,and the related polymorphic loci were screened by direct sequencing and its correlation with milk quality traits was analyzed.At the same time,the haplotypes were obtained by linkage disequilibrium analysis using Haploview 4.2software,and the association of PDE10Agene haplotypes with milk quality traits in dairy cows was analyzed.The results showed that three SNPs were detected in introns 12and 13of PDE10Agene,among which I12-3228A>G and I13-13A>G loci were significantly associated with milk yield,milk fat,milk protein,milk sugar,somatic cell count,dry matter content and corrected milk yield in Chinese Holstein cows(P<0.05).The results of linkage analysis revealed that H1H3genotype was found to have higher milk yield and higher milk fat,milk protein and lactose contents.The results of this study suggest that the PDE10Agene can be used as a candidate gene affecting milk production traits in Chinese Holstein cows for marker-assisted selection in animal breeding.
