摘要
目的以发生侧颈淋巴结转移的甲状腺乳头状癌(PTC)为研讨对象,旨在找到阻断淋巴管形成、降低侧颈淋巴结转移率的方法。方法长链非编码RNA(lncRNA)测序得出,在发生侧颈淋巴结转移的PTC中存在差异表达基因,筛选明显上调、下调的基因。结果发生侧颈淋巴结转移的PTC中有4782个上调lncRNA,3807个下调lncRNA。结论lncRNA H19、lncRNA CHI3L1出现明显表达差异,推测与发生侧颈淋巴结转移有关,可作为靶点构建lncRNA-X敲除慢病毒,验证是否与淋巴管形成有关。
Objective With regard to papillary thyroid carcinoma(PTC)with lateral-neck lymph node metastasis,find a method to block the formation of lymphatic vessels and reduce the rate of lateral neck lymph node metastasis.Methods Long non-coding RNA(lncRNA)sequencing showed that there were differentially expressed genes in PTC with lateral cervical lymph node metastasis,and the significantly up-regulated and down-regulated genes were screened.Results There were 4782 up-regulated lncRNAs and 3807 down-regulated lncRNAs in PTCs with lateral cervical lymph node metastasis.Conclusion There are significant differences in the expression of lncRNA H19 and lncRNA CHI3L1,which are presumed to be related to the metastasis of lateral cervical lymph node.They can be used as targets to construct lncRNA-X knockout lentivirus to verify whether it is related to lymphatic vessel formation.
