摘要
目的:利用ISSR技术对穿心莲四倍体8个株系和国内外25个不同产地穿心莲进行了遗传多样性分析,为穿心莲国内外地区引种、优良品种的选育和品质改良提供科学依据。方法:用CTAB法提取穿心莲基因组DNA,筛选出8条ISSR引物对33份穿心莲材料进行遗传分析。采用UPGMA法对穿心莲样品进行遗传多样性和聚类分析。结果:33份穿心莲样品共扩增出62条条带,其中有31条多态性条带,多态性条带比率为50%。国内各个产地之间遗传多样性较低,国内与国外(美国和加拿大)产地的穿心莲材料相比遗传差异性较大。结论:穿心莲四倍体与二倍体之间具有一定的遗传差异性,国外穿心莲种质资源遗传多样性较高,该结果为穿心莲良种选育及提高穿心莲品质积累了必要的实验数据。
Objective:To analysis the genetic diversity of eight Andrographis paniculata tetraploid lines and 25 Andrographis paniculata from different habitats at home and abroad by ISSR technique,so as to provide scientific basis for the introduction,selection and breeding of superior varieties and quality improvement of Andrographis paniculata at home and abroad.Methods:Genomic DNA of Andrographis paniculata was extracted by CTAB method,and 8 ISSR primers were screened out for genetic analysis of 33 Andrographis paniculata materials.UPGMA was used for the genetic diversity analysis and cluster analysis of Andrographis paniculata.Results:A total of 62 bands were amplified from 33 Andrographis paniculata samples,of which 31 bands were polymorphic,the percentage of polymorphic banding was 50%.The genetic diversity among different producing areas in China was low,and the genetic difference between Chinese and abroad(United States and Canada)was large.Conclusion:There are genetic differences between tetraploid lines and diploid lines of Andrographis paniculata,and the genetic diversity of abroad Andrographis paniculata germplasm resources is high.The results lay necessary experimentd basis for the selection and breeding of superior varieties and the improvement of Andrographis paniculata quality.
