摘要
目的:建立同时测定女贞子中酪醇、红景天苷、羟基酪醇、女贞酸、木樨榄苷-11-甲酯、女贞次苷、特女贞苷含量的方法,研究女贞子酒蒸前后活性成分在大鼠主要器官组织中的分布,探讨女贞子炮制机制和药效的相关性。方法:分别灌胃给予SD大鼠女贞子和酒女贞子提取物,采用UHPLC-MS/MS法测定大鼠组织(心、肝、脾、肺、肾、胃、小肠、脑)中7个成分的含量,探究酒蒸对女贞子中活性成分在大鼠体内组织分布的影响。结果:女贞子酒蒸能明显改变7个成分在大鼠体内的分布,酒蒸后酪醇、红景天苷、羟基酪醇、女贞酸在大鼠部分组织中的浓度呈升高趋势,木樨榄苷-11-甲酯、女贞次苷、特女贞苷在大鼠部分组织中的浓度呈降低趋势。结论:酒蒸对女贞子的活性成分在大鼠体内的组织分布有较大影响,该研究结果为探讨女贞子炮制机制提供了实验依据。
Objective:To establish a method for simultaneous determination of tyrosol,salidroside,hydroxytyrosol,nuezhenidic acid,oleoside-11-methyl ester,1-O-β-D-glucosylformoside and specnuezhenide in Ligustri Lucidi Fructus,and to study the distribution of active compounds of Ligustri Lucidi Fructus before and after wine steaming in main organs and tissues of rats,and to explore the correlation between processing mechanism of Ligustri Lucidi Fructus and efficacy.Methods:SD rats were given the extracts of Ligustri Lucidi Fructus and wine steamed Ligustri Lucidi Fructus by intragastric administration,respectively.The contents of 7 components of Ligustri Lucidi Fructus in tissues(heart,liver,spleen,lung,kidney,stomach,small intestine and brain)were determined by UHPLC-MS/MS to explore the effect of wine steaming on the distribution of active components of Ligustri Lucidi Fructus in rats.Results:Wine processing could significantly change the distribution of 7 components of Ligustri Lucidi Fructus in rats,and the concentrations of tyrosol,salidroside,hydroxytyrosol and nuezhenidic acid in the some tissues of rats increased after wine processing.The concentration of oleoside-11-methyl ester,1‴-O-β-D-glucosylformoside and specnuezhenide in the some tissues of rats decreased.Conclusion:Wine processing has a great effect on the tissue distribution of active components of Ligustri Lucidi Fructus in rats.The results provide an experimental basis for exploring the processing mechanism of Ligustri Lucidi Fructus.
