摘要
目的:通过研究麦冬去心前后化学成分变化情况,初步分析麦冬“去心”的科学性。方法:采用HPLC法建立麦冬(MDM)、去心麦冬(MDP)及麦冬心(MDC)指纹图谱,通过色谱峰指认结合相似度评价、主成分分析(PCA)和偏最小二乘判别分析(PLS-DA)探讨3种样品间的差异;同时测定3种样品中浸出物、总皂苷、总多糖、甲基麦冬黄烷酮A及甲基麦冬黄烷酮B的含量以充分评价麦冬去心前后的差异性。结果:建立了3种样品HPLC指纹图谱,分别确定了16个共有峰,相似度均大于0.940。PCA和PLS-DA将30批样品分为3类,筛选出6个质量差异标志物。含量测定结果显示MDC除总多糖与浸出物外各成分含量均显著高于MDM和MDP。结论:麦冬心中化学成分含量较高,但在药材中所占比例较小,且“去心”处理操作不便,故在实际应用时,建议可不做“去心”处理。
Objective:To study the changes of chemical compositions before and after Radix Ophiopogonis processing and to analyze the scientific of whether Radix Ophiopogonis should removal the core.Methods:The fingerprints of the Radix Ophiopogonis(MDM),"core removal"of Radix Ophiopogonis(MDP)and Radix Ophiopogonis core(MDC)were established by HPLC,respectively.The differences among 3 kind of samples were discussed by chromatographic peak identification combined with similarity evaluation,principal component analysis(PCA)and partial least square discriminant analysis(PLS-DA).In addition,the contents of water extract,total saponins,total polysaccharides,methylophiopogonanone A,methylophiopogonanone B in 3 kind of samples were determined to fully perform variance analysis.Results:The HPLC fingerprints of 3 kind of samples were established,and 16 common peaks were determined,respectively.The similarity of the fingerprints was more than 0.940.According to PCA and PLS-DA,30 batches of samples were divided into 3 categories and 6 differential components were screened out.The results of content determination showed that the contents in MDC were significantly higher than those in MDM and MDP except total polysaccharides and water extract.Conclusion:The contents of chemical components in Radix Ophiopogonis core are high,but the proportion in medicinal materials is small,and the operation of"removing core"is inconvenient,so it is suggested not to do"removing core"treatment in practical application.
