摘要
目的 通过对苦豆总碱、苦参总碱的体外抗柯萨奇B组 3型病毒 (CVB3 )作用的对比研究 ,为其在临床应用提供实验依据。方法 以盐酸胍作为实验阳性对照药物 ,采用细胞培养技术及细胞病变效应 (CPE)抑制法、噻唑蓝 (MTT)比色法、组织培养半数感染 (TCID50 )微量法进行苦豆总碱、苦参总碱的体外抗CVB3 作用研究。结果 苦豆总碱、苦参总碱均有明显的抗CVB3 作用 (P <0 .0 5 ) ,对CVB3 的抑制指数均 >2 ,细胞保护率最高可达 95 % ,其中感染后给药及药物与病毒先作用 2小时然后加入细胞层的给药方法抗病毒效应较强。结论 苦豆总碱、苦参总碱在体外有明显抗CVB3 。
Objectives To study the role of total alkaloids of Sophora alopecuroides and Sophora flavescens in anti-coxsackievirus B 3 in vitro.Methods The role of Sophora Alopecuroides L. and Sophora flavescens Alkaloids in anti-coxsackievirus B 3 in vitro was detected by cell culture technology, cytopathic effect inhibition, MTT colormetric assay and 50% tissue culture infection dose microassay, meanwhile,Guanidine HCl was used as positive control.Results Total alkloids of Sophora Alopecuroides L.and Sophora flavescens showed remarkably anti-CVB 3 effect and inhibited significantly cytopathogensis index of CVB 3, the highest protection rate was 95%, the role in Ⅱgroup (drugs administered after infection with CVB 3) and Ⅳ group (drugs and virus simultaneously administered after being co-incubated for two hours ) were better than those of other groups in vitro.Conclusions Total Alkaloids of Sophora Alopecuroides L. and Sophora flavescens showed significantly the anti-CVB 3 effect and inhibited cytopathogensis index of CVB 3, the mechanism of anti-virus effect exerted by sophocarpine seems likely to kill the virus directly and/or to interfere with replication of virus indirectly.
