摘要
目的探讨枸杞多糖(LBP)对2,4-二氯苯氧乙酸(2,4-D)染毒初断乳大鼠神经行为和海马神经元细胞凋亡的保护作用。方法SPF级初断乳SD大鼠48只,雌雄各半,随机分为Control组、2,4-D组、LBP组和2,4-D+LBP组各12只,旷场实验、高架十字迷宫观察实验大鼠行为学变化,试剂盒测定大鼠血清抗氧化酶活性,HE染色观察大鼠海马CA3区锥体细胞的形态,TUNEL荧光染色法观察海马CA3区神经元凋亡,免疫组织化学方法检测海马组织凋亡蛋白Bcl-2和Bax的表达。结果(1)2,4-D组大鼠脑重和脑脏器系数降低,大鼠血清中超氧化物歧化酶(SOD)和谷胱甘肽过氧化物酶(GSH-Px)活性降低,丙二醛(MDA)含量升高(P均<0.01);海马CA3区锥体细胞数减少,光镜下观察细胞间隙增宽,排列不整齐,细胞与周围组织联系疏松,部分胞核固缩呈三角形或不规则形;大鼠中央格停留时间延长,穿行格数、直立次数和修饰时间减少,进入开放臂次数减少,开放臂停留时间缩短,开放臂停留时间百分比降低(P均<0.05);(2)与2,4-D组比较,2,4-D+LBP组大鼠脑重和脑脏器系数增加,血清SOD和GSH-Px活性升高,MDA含量降低(P均<0.01);海马CA3区锥体细胞数量较2,4-D组增多,部分细胞恢复正常形态(P均<0.01);中央格停留时间缩短,穿行格数、直立次数和修饰时间均延长,进入开放臂次数增多,开放臂停留时间延长,开放臂停留时间百分比升高(P均<0.05);(3)TUNEL染色结果显示,2,4-D组大鼠海马神经元阳性细胞数增多,细胞凋亡率升高(P均<0.01),2,4-D+LBP组凋亡细胞数少于2,4-D组,呈现绿色或淡绿色荧光;(4)免疫组织化学方法发现2,4-D组大鼠海马组织Bcl-2表达降低,Bax表达升高,Bcl-2/Bax比值降低(P均<0.05),2,4-D+LBP组大鼠海马CA3区Bcl-2表达升高,Bax表达降低,Bcl-2/Bax比值升高(P均<0.05),神经元染色以胞浆有棕黄色物质沉积为主,也有胞膜或核膜染色。结论2,4-D染毒大鼠表现出抑郁和焦虑状态,大鼠血�
Objective To investigate the protective effects of lycium barbarum polysaccharide(LBP)on neural behavior and apoptosis of hippocampal neurons in 2,4-dichlorophenoxyacetic acid(2,4-D)infected and newly weaned rats.Methods 48 SPF SD rats were randomly divided into control group,2,4-D exposure group,LBP group and 2,4-D+LBP group,12 in each group.The behavior changes of the experimental rats were observed by open field experiment and elevated cross maze.The serum antioxidant enzyme activity of the rats was determined by kit,and the morphology of pyramidal cells in hippocampus CA3 region of rats was observed by HE staining.The neuronal apoptosis in hippocampal CA3 region was observed by TUNEL fluorescence staining,and the expression of apoptotic proteins Bcl-2 and Bax in hippocampal tissues was detected by immunohistochemistry.Results(1)The brain weight and brain organ coefficient of rats exposed to 2,4-D decreased,the activities of SOD and GSH-Px decreased,and the content of MDA increased(P all<0.01).The number of pyramidal cells in hippocampal CA3 region decreased,and the space between cells was widened and arranged irregularly under light microscope.The connection between cells and surrounding tissues was loose,and some nuclei were pyknotic in triangular or irregular shape.The retention time of central lattice was prolonged,and the number of crossing lattice,upright frequency and modification time were decreased,and the number of entering open arm was decreased,and the retention time of open arm was shortened,and the percentage of retention time of open arm was decreased(P all<0.05).(2)Compared with 2,4-D group,the brain weight and cerebral organ coefficient of 2,4-D+LBP group increased,the activities of SOD and GSH-Px in serum increased,and the content of MDA decreased(P all<0.01).The number of pyramidal cells in hippocampal CA3 region was higher than that in 2,4-D group,and some of them returned to normal form(P all<0.01).The retention time of central lattice was shortened,the number of moving through the lattice
