摘要
目的:探讨紫龙金片对S180肉瘤小鼠的抑瘤作用及其作用机制。方法:选择10只未接种瘤细胞的小鼠定为正常组,将确定造模成功的40只荷瘤小鼠随机分为荷瘤组、紫龙金组、化疗组、紫龙金+化疗组,每组各10只。荷瘤组给予0. 2 m L蒸馏水灌胃,0. 2 m L生理盐水标准腹腔注射;紫龙金组给予紫龙金溶剂水1. 3 g·kg-1灌胃,0. 2 m L生理盐水标准腹腔注射;化疗组给予0. 2 m L蒸馏水灌胃,52 mg·kg-1环磷酰胺水溶液标准腹腔注射;紫龙金+化疗组给予紫龙金溶剂水1. 3 g·kg-1灌胃,52 mg·kg-1环磷酰胺水溶液标准腹腔注射;正常组不给药。称取各组小鼠的瘤质量,并计算抑瘤率。利用流式细胞仪检测S180肉瘤小鼠外周血中的Treg细胞及Th1、Th2、Th17所分泌的细胞因子白细胞介素-4(interleukin-4,IL-4)、IL-6、IL-10、IL-2、IL-17A、干扰素-γ(interferon-γ,IFN-γ)的含量。结果:紫龙金组、化疗组和紫龙金+化疗组抑瘤率分别为10. 24%、16. 98%、22. 10%,其中两药联用组抑瘤率最高、瘤质量最小。紫龙金片能调节S180肉瘤小鼠外周血中Treg细胞,使其与正常组比较,差异无统计学意义(P> 0. 05)。瘤组与正常组比较,IL-2、TNF和IFN-γ水平均有所升高,差异具有统计学意义(P <0. 05)。与正常组比较,荷瘤组IL-4、IL-6明显下降,差异具有统计学意义(P <0. 05)。与荷瘤组比较,化疗组、紫龙金组及紫龙金+化疗组IL-4水平差异无统计学意义(P> 0. 05);与荷瘤组比较,紫龙金组IL-6水平提高,差异具有统计学意义(P <0. 05)。紫龙金及紫龙金+化疗组能降低IL-10及IL-17水平,但与正常组比较,差异无统计学意义(P>0. 05)。结论:紫龙金片可通过调节肿瘤机体免疫相关细胞因子水平来促进细胞及体液的免疫应答,其抑瘤作用机制与对Th1、Th2、Th17、Treg细胞因子免疫格局的调控密切相关。
Objective: To investigate the antitumor effect and mechanism of Zilongjin Tablets(ZLJT) on S180 sarcoma mice. Methods: Ten mice without inoculated tumor cells were selected into the normal group,and 40 mice with tumor due to modeling were randomly divided into the tumor-bearing group,the ZLJT group,the chemotherapy group,and the ZLJT + chemotherapy group,with10 mice in each group. The ones in the tumor-bearing group were given 0. 2 m L distilled water for gavage,and 0. 2 m L normal saline was injected intraperitoneally. The ones in the ZLJT group were given ZLJT solvent1. 3 g·kg^-1 for intragastric injection,0. 2 m L normal saline standard intraperitoneal injection. The ones in the chemotherapy group were given 0. 2 m L distilled water for intragastric injection,52 mg·kg^-1 cyclophosphamide water for standard intraperitoneal injection. The ones in the ZLJT + chemotherapy group were given ZLJT solvent 1. 3 g·kg^-1 for intragastric administration,52 mg·kg^-1 cyclophosphamide solution for standard intraperitoneal injection,and the ones in the normal group did not receive drugs. The tumor mass of each group was calculated and the tumor inhibition rate was calculated. The contents of Treg cells and cytokines interleukin-4(IL-4),IL-6,IL-10,IL-2,IL-17 A and interferon-gamma(IFN-gamma) secreted by Th1/Th2/Th17 in peripheral blood of S180 sarcoma mice were detected by flow cytometry. Results: The tumor inhibition rates of the ZLJT group,the chemotherapy group,and the ZLJT + chemotherapy group were 10. 24%,16. 98% and 22. 10%,respectively. The combination of two drugs had the highest tumor inhibition rate and the lowest tumor mass. ZLJT could regulate Treg cells in peripheral blood of S180 sarcoma mice,and there was no significant difference between the above three groups and the normal group(P〈0. 05). The levels of IL-2,TNF and IFN-gamma in the tumor-bearing group were higher than those in the normal group,and the differences were statistically significant(P〈0. 05).Compared with the
