摘要 目的:探讨二仙汤水提物对小鼠脾脏细胞和卵巢颗粒细胞作用的影响。方法:取小鼠脾脏和卵巢分别制备脾单个核细胞和卵巢颗粒细胞体外培养,加入二仙汤水提物,MTT法测细胞增殖情况;ELISA法测细胞上清液中IL-1、IL-5、IFN-γ以及雌二醇(E2)的浓度;荧光实时定量PCR检测雌激素受体(ERα、ERβ)及IL-1受体1(IL-1R1)的m RNA表达。结果:二仙汤对小鼠脾单个核细胞及卵巢颗粒细胞均具有较好的促增殖作用(P<0.01或P<0.05);可提高脾细胞培养上清中IL-1、IFN-γ及IL-5的含量(P<0.01或P<0.05),提高卵巢颗粒细胞上清中E2水平(P<0.05),使水提二仙汤组的Th1/Th2比值显著降低(P<0.05);并诱导脾细胞上ERα的m RNA表达(P<0.05)。结论:二仙汤水提物对小鼠免疫和内分泌功能均有较好的促进作用,可诱导Th1/Th2平衡向Th2偏移。 Objective: To investigate the effects of water extract of Erxian Decoction on spleen and ovarian function in mice. Methods: Cultivate the mouse splenic cells and ovarian granulosa cells and add water extract of Erxian Decoction, respectively. MTT assay was used to evaluate the cell proliferation. Enzyme linked im- munosorbent assay was used to observe the concentration of IL-1, IL-5 and IFN-y as well as E2 in the super- natant of cells. The mRNA levels of ERα, ERβ and IL-1R1 were validated by real-time quantitative PCR. Re- suits: Erxian Decoction promoted the proliferation of ovarian granulosa cells and splenic cells (P〈0.05 or P〈0.01). Erxian Decoction can increase the concentration of IL-1, IL-5 and IFN-3, in the supernatant of splenic cells in mice (P〈0.05 or P〈0.01), and increase the concentration of E2 in the supernatant of ovarian granulosa cells (P〈0.05). Water extract of Erxian Decoction can decrease the ration of Thl/Th2 and increase the lever of ERa mRNA on splenic cells (P〉0.05). Conclusion: Water extract of Erxian Decoction has better effects on regulating splenic cells and ovarian granulosa cells, and can induce the balance of Thl/Th2 to Th2.
