摘要
为筛选高生物活性的益生菌菌株,通过热处理从健康猪肠道内容物、粪便、土壤等来源筛选具有较高抗逆性的菌株,综合形态特征、生理生化特性及16S r DNA序列分析进行鉴定;通过耐人工胃液、耐人工肠液、耐胆盐、耐高温、产酶试验、药敏试验、抑菌试验来研究菌株的特性。综合鉴定菌株ZZS16-3为枯草芽孢杆菌(Bacillus subtilis),其在p H为2.0的人工胃液中,存活率可达到65.8%,在胆盐浓度为0.3%时存活率为51.2%,在100℃处理3 min后其存活率达到11.2%,可产生蛋白酶、淀粉酶、脂肪酶等,对于绝大多数的药物是敏感的,对金黄色葡萄球菌则有明显的抑菌效果。枯草芽孢杆菌ZZS16-3具有较强的抗逆性和潜在的益生性能,为研制微生态制剂提供了优良的菌种参考。
To screen probiotic strains with high biological activities.Heat treatment was used to screen high resistance strains from healthy pigs' intestinal contents ,feces, and soil around the pig farms.The strains were i- dentified by morphological,physiological and biochemical characteristics and 16S rDNA sequence analysis.The characteristics of the strains were studied by testing their resistance to artificial gastric juice, artificial intestinal fluid ,bile salts and high temperature.Enzyme production testing,susceptibility testing and antimicrobial testing were also conducted.A Swine-origin strain ZZS16-3 was comprehensively identified as Bacillus subtilis. Its sur- vival rate was up to 65.8% in the artificial gastric juice at pH 2.0,51.2% in the bile salt concentration of 0.3%, 11.2% after the treatment at 100 ℃ for 3 min.h produced protease, amylase ,lipase and so on, and it was sensitive to the majority of antibiotics.It showed significant inhibitory effect on Staphylococcus aureus.It was in- dicated that Bacillus subtilis ZZS16-3 had strong resistance to the environment and potential prebiotic proper- ties.Thus an excellent strain was provided to the development of prebiotics.
