摘要
目的研究丹参酮ⅡA对缺血再灌注损伤大鼠心肌组织和心律失常情况的保护作用及其对心肌组织缺血修饰白蛋白(IMA)和心型脂肪酸结合蛋白(H-FABP)的影响。方法取Wistar大鼠40只,随机分成5组,每组8只,即假手术组、模型组和丹参酮ⅡA高、中、低剂量组(20,10,5mg·kg-1·d-1),假手术组和模型组给予等容量生理盐水,丹参酮ⅡA各剂量组给予相应药物,均为灌胃给药,喂养7d,末次给药2h后进行手术。除假手术组在手术时只穿线不进行冠脉结扎和再灌注之外,其余各组采用冠脉结扎再松开的办法制备大鼠缺血再灌注损伤模型。记录Ⅱ导联心电图,并对大鼠心律失常进行评分;常规HE染色观察心肌组织的病理改变;ELISA法检测心肌组织IMA、H-FABP的表达。结果大鼠心肌缺血再灌注损伤后,心肌组织病理损伤心律失常发生明显,心肌IMA含量显著增加,H-FABP含量显著降低。丹参酮ⅡA干预后,心律失常评分显著降低,心肌组织病理改变减轻,心肌组织I-MA含量显著降低、H-FABP水平明显升高,丹参酮ⅡA高、中剂量组心肌组织IMA含量,与模型组比较,具有显著性差异(P<0.05);丹参酮ⅡA高剂量组H-FABP含量显著增加,与模型组比较,具有显著性差异(P<0.05)。结论丹参酮ⅡA对缺血再灌注损伤大鼠心肌组织和心律失常情况具有良好的保护作用,可抑制缺血再灌注损伤后大鼠心肌组织IMA的升高和H-FABP水平的降低。
Objective To observe the effect of tanshinone ⅡA (T ⅡA ) on arrhythmia and levels of ischemia modified albumin (IMA) and heart type- fatty acid binding protein (H -FABP) in rats with myocardial ischemia- reperfusion injury. Methods Forty Wistar rats were equally randomized into 5 groups: pseudo- operation group (normal saline), model group (normal saline), and high - , middle - and low -dose T ⅡA groups (in the dose of 20, 10 and 5 mg ·kg^-1·d^-1 respectively). The rats were treated for 7 days, and received operation 2 hours after the last medication. Rats in pseudo - operation group received threading without ligation, while animal model of myocardial ischemia - reperfusion injury was established by ligating the left - anterior coronary artery for 15 minutes and then loosening for 30 minutes. During the operation, electrocardiogram was traced continuously to observe the arrhythmia. HE staining was used to observe the pathological changes of myocardium. Levels of IMA and H -FABP in myocardium were determined by enzyme -linked immunosorbent assay (ELISA). Results Ischemic changes of the myocardium and arrhythmia occurred, myocardial IMA level increased and H - FABP level decreased in the model rats. Tanshinone ⅡA obviously attenuated arrhythmia caused by ischemia - reperfusion injury ( P 〈 0.05), relieved the pathology changes in myocardium, decreased IMA level and increased H - FABP level. The difference of IMA level between high - and middle - dose T ⅡA groups and the model group was significant ( P 〈 0.05). So did the difference of H - FABP level between high - dose T ⅡA group and the model group ( P 〈 0.05). Conclusion Tanshinone ⅡA has a protective effect against arrhythmia in rats with myocardial ischemia - reperfusion injury, which can inhibit the increase of IMA and the decrease of H - FABP after the injury.
