摘要
血脑屏障的存在使得几乎所有的大分子治疗因子和超过98%的小分子药物难以到达脑组织发挥其治疗作用。因此,探索能高效传递药物和生物大分子进入中枢神经系统进行相应治疗的模式成为当前的重要课题。新近研究证实:免疫脂质体能安全有效地介导药物穿透血脑屏障。本文通过综述国内外关于跨血脑屏障给药技术的最新研究进展,对血脑屏障的结构及调控、免疫脂质体的性质特点、在脑组织内的释放机制及给药途径进行总结,并对其临床应用做出展望。
The blood-brain battier excludes from the brain 100% of large-molecule therapeutic factor and more than 98 % of small-molecule drugs. Therefore, discovery of new modalities for effective delivery of drugs and biomacromoleeules to the central nervous system is of great need and importance for treatment of neuropathy. The present investigations provid successful applications of immunoliposome technology is the safe and effective way of deliverying drags across the blood-brain barrier. In this paper, the structure and regulation of BBB are expounded, the characteristic, mechanisms of drug release and the route of administration are reviewed. In addition, prospects of future uses of immunoliposome-based drug delivery systems and clinical trials are discussed.
