基质金属蛋白酶-26和唾液酸化Lewis-X蛋白在乳腺癌中的表达及其意义 被引量:1

摘要 目的研究基质金属蛋白酶-26(matrix metalloproteinase-26,MMP-26)和唾液酸化Lewis-X(sialy lLewis-X,SLeX)抗原蛋白在乳腺癌组织中的表达及与淋巴结转移的关系。方法应用免疫组化催化信号放大法检测120例乳腺癌、71例转移淋巴结和30例正常乳腺组织中MMP-26和SLeX蛋白的表达水平。结果MMP-26和SLeX蛋白在正常乳腺组织上皮细胞中的表达阳性率分别为16.7%(5/30)和20.0%(6/30),而在乳腺癌组织中两者的表达阳性率分别为63.3%(76/120)和86.7%(104/120),差异有统计学意义(P<0.05)。在有淋巴结转移的乳腺癌组织中,SLeX表达阳性率〔100%(71/71)〕明显高于无淋巴结转移者〔67.3%(33/49)〕,P<0.05,而MMP-26表达阳性率差异则无统计学意义(P>0.05)。在有癌转移的淋巴结中,MMP-26和SLeX蛋白表达阳性率分别为23.9%(17/71)和88.7%(63/71),后者与原发癌组织中的表达具有较高一致性(r=0.874,P<0.05)。结论乳腺癌组织中MMP-26和SLeX蛋白均呈高水平表达,并且后者与淋巴结转移密切相关。作为预测乳腺癌侵袭和转移的肿瘤相关抗原,联合检测MMP-26和SLeX对评估患者的预后具有重要实用价值。 Objective To investigate the expressions of matrix metalloproteinase-26 (MMP-26) and sialyl Lewis-X (SLeX) in breast carcinoma and their relation with lymphaden metastasis. Methods The expressions of MMP-26 and SLeX protein in breast carcinoma (n=120), metastatic lymph node (n=71) and normal mammary gland tissue (n= 30) were studied by catalyzed signal amplification (CSA) immunohistochemical method. Results The positive rate of MMP-26 and SLeX protein expression were 16.7 % (5/30) and 20.0 % (6/30) in normal breast tissues, and 63.3% (76/120) and 86.7% (104/120) in breast carcinoma, resepectively. The expressions of MMP-26 and SLeX protein in breast carcinoma tissues were significantly higher than those in remote normal breast tissues (P〈0.05). The positive rate of SLeX protein in tissues of breast carcinoma with lymphaden metastasis was higher than that without lymphaden metastasis (P〈0.05). There was no difference of MMP-26 protein expression in tissues of breast carcinoma between the patients with lymphaden metastasis and without lymphaden metastasis (P〉0.05). The positive rate of MMP-26 and SLeX protein expression were 23.9%(17/71) and 88.7%(63/71) in metastatic lymph node, resepectively. The expression of SLeX protein in metastatic lymph node was correlated with that in tissues of breast carcinoma (r=0. 874,P〈0.05). Conclusion MMP-26 and SLeX protein are highly expressed by breast carcinoma tissues. SLeX is closely related to lymphaden metastasis. MMP-26 and SLeX may serve as tumor-associated antigen for estimating invasion, metastasis and prognosis in breast carcinoma.
