摘要
目的:响应面分析法(RSM)优化银离子络合法提取鱼油乙酯中EPA及DHA的最佳工艺。方法:在单因素试验的基础上,依据Box-Benhnken的中心组合试验设计原理,选取对(EPA+DHA)转移率影响显著的3个因素(硝酸银的浓度、料液比、解离温度),采用3因素3水平的响应面分析方法,对工艺条件进行优化。结果:该3因素对转移率存在显著的相关性,最佳工艺条件为硝酸银浓度4.9 mol/L、络合温度0℃、料液比1∶3.76、解离温度51℃、解离时间30 min,(EPA+DHA)的最高理论转移率为62.80%。结论:响应面法优化得到的工艺参数可信,正确可行。
Objective: To seek the optimum technology of extracting EPA and DHA in fish oil with silver ion complexation by response surface methodology(RSM). Method: Based on the results of single factor tests, according to Box-Behnken factorial design principle, the 3 factors of the concentration of AgNO3 solution, ratio of AgNO3 solution to fish oil and dissociation temperature were selected as the obvious factors and RSM with three factors and three levels was used to optimize the extraction conditions for the yield of EPA and DHA. Result: The response surface analysis showed that the three factors had significant correlation to EPA and DHA. The optimum extraction conditions for EPA and DHA were as follows: concentration of aqueous AgNOs is solution 4.9 mol/L, complexation temperature is 0 ℃, ratio of fish oil to AgNOs solution is 1:3:76, dissociation temperature is 51 ℃ and dissociation time is 30 min. And that the conversion of EAP and DHA was up to 62.80% of theoretical value. Conclusion: The extraction conditions parameter obtained by RSM is credible.
