柴黄益肾方对嘌呤霉素氨基核苷肾病大鼠尿蛋白及纤溶功能的影响 被引量:3

摘要 目的探讨柴黄益肾方提取物(CYE)对嘌呤霉素氨基核苷(PAN)肾病大鼠尿蛋白及纤溶功能的影响。方法大鼠经颈静脉一次性注射PAN5mg/100g体重制备大鼠肾病模型,于造模后第二天开始灌胃给药,连续给药21周后检测24小时尿蛋白含量,腹主动脉取血测定TPA、PAI、AT-Ⅲ纤溶功能。结果21周给药蒙诺与CYE均能显著降低尿蛋白,升高组织型纤溶酶原激活物活性(TPA),降低纤溶酶原活化物抑制因子(PAI),增强抗凝血酶Ⅲ(AT-Ⅲ)活性。结论柴黄益肾方提取物能抑制嘌呤霉素氨基核苷诱导的大鼠肾病模型的尿蛋白排泄,并能改善纤溶功能。 Objective: To investigate the effects of Chaihuang-Yishen Formula Extract (CYE)on urinary protein and the function of fibrinolysis of rats with nephrosis induced by puromycin aminonueleoside (PAN). Methods: Wistar rats were randomly divided into six groups ; sham operation, PAN-model , PAN-model with high dosage CYE (2.2g/kg · d) , PAN-model with median-dosage CYE (1.1g/kg · d) ,PAN-model with low-dosage CYE (0.55g/kg · d) , and PAN-model with Fosinopri(FP). The PAN-model was induced by jugular vein injection of PAN at a dose of 5mg/100g body weight. Quantities of 24 hours urinary protein excretion and the function of fibrinolysis were examed when all rats were sacrificed after 21 weeks. Results: Treatment of CYE and FP significantly reduced protein excretion and change the function of fibrinol- ysis such as Tissue-type plasminogen activator (TPA) , Plasminogen activator(PAI) and Antithrombin type Ⅲ (AtⅢ). Conclusion: Chinese herbal Formula-Chaihuang-Yishen-Extract can reduce the urinary protein excretion and can regulate the changes of fibrinolysis fuetion of rats with nephrosis induced by puromycin aminonucleoside.
