石上柏和蒲葵子对蛋白激酶C活性的影响 被引量:17

摘要 采用DE52柱层析法部分纯化大鼠中的蛋白激酶C,研究了石上柏和蒲葵子醇提物对蛋白激酶C活性的影响。为了证明测定系统的可靠性,观察了多粘菌素B(已知蛋白激酶C抑制剂)对蛋白激酶C活性的影响。表明:石上柏醇提物对蛋白激酶C有强烈的抑制作用,IC502.2μg/ml;100μg/ml的蒲葵子醇提物对蛋白激酶C的抑制率为66.6%,降低药物浓度抑制作用减弱;多粘菌素B对蛋白激酶C有抑制作用,IC5017.7U/ml。 Protein Kinase C (PKC ) was partially purified from rat brain by DEAE-52 column chromatography, and the effects of ethanol extracts of Selaginella doederleinii and Livistona chinensis on PKC activity were investigated.In order to verify the reliability of the assay system, the effect of polymyxin B, a known PKC inhibitor, on the activity of PKC was also tested. Results showed that the ethanol extract of S. doederleinii strongly inhibited PKC activity with IC50 2.2μg/ml. The ethanol extract of L. chinensis (100μg/ml ) inhibited PKC activity by 66.6%. Polymyxin B inhibited PKC activity with IC5017.7 U/ml.
