摘要
采用乙型脑炎重组痘苗病毒J1和J3株,两者均含乙脑病毒(JEV)的E、NS1和NS2A基因,后者还含PrM基因,在BHK-21-15细胞中培养表达,经免疫荧光法(IFA)测定,两者均可高效表达JEV特异性抗原。用该重组病毒免疫瑞士种小鼠,经IFA检测证明J1、J3均可诱生抗JEV抗体,其中J3免疫鼠还产生中和抗体(N)和血抑抗体(HI),且其抗JEV攻击能力显著增高。
he Third Military Medical University)Two JEV-vaccinia recombinants J1 and J3 were engineered for expression of the JEVantigens. Both J1 and J3 contained the E,NS1 and NS2A genes,J3 had the PrM gene inaddition to the above three genes. Indirect immunofluorescent assay revealed that J1 and J3expressed JEV antigens in recombinant-infected BHK cells. Immunization of 15-day-oldmice with the recombinants resulted in immune response to JEV. Mice vaccinated with J3produced both hemagglutinin-inhibiting and virus-neutralizing antibodies to JEV and showedincreased resistance to JEV chanllenge;while J1 had no such effects.Therefore PrM gene of J3played an important role in inducing protective immune response.
