摘要
目的从分子生化水平上对田间农艺性状表现差异的6种类型盾叶薯蓣进行比较分析。方法采用垂直板聚丙烯酰胺凝胶电泳技术,对6种类型盾叶薯蓣的叶片进行过氧化物酶(POD)同工酶测定。结果POD同工酶在不同类型盾叶薯蓣间有所不同。根据酶带的分离结果,可将此6种类型大致划分为三大类,即有花纹大三角叶型(A)与无花纹大三角叶型(B)为一类;有花纹小三角间心形叶型(E)与无花纹心形叶型(D)、无花纹小三角间心形叶型(F)为一类;有花纹心形叶型(C)为单独一类。结论根据酶带分布差异的分析结果,可以初步对形态表现不同的盾叶薯蓣加以归类。同工酶电泳分析方法可作为薯蓣选种研究的辅助工具。
Objective To analyze the differences of six different types of Dioscorea zingiberendid C.H.Wright on the molecule biochemistry level.Methods The POD isoenzyme analysis in leaves on six different types of Dioscorea zingiberendid C.H.Wright was carried out with vertical slab polyacrylamide gel electrophoresis technique.Results The results indicated:six types of Dioscorea zingiberendid C.H.Wright can be compartmentalized 3 groups,A and B can be considered as a genre,D and E,F can be considered as another genre,and C can be considered as a separate genre.Conclusion From the analysis of the POD isoenzyme belts distributing,we can categorize the six different types preliminarily.And the technique of isoenzyme analysis can be applied in the seed selecting studies of Dioscorea zingiberendid C.H.Wright.
