摘要
目的探讨唾液乳杆菌对类风湿性关节炎(RA)的治疗效果及作用。方法选择牛Ⅱ型胶原为免疫原,对雄性SD大鼠建立胶原诱导性关节炎(CIA)模型。随机选取24只造模成功的大鼠,随机分成CIA模型组、唾液乳杆菌组、甲氨蝶呤组,每组8只,另外选择8只大鼠作为健康组。健康组和CIA模型组均给予细菌培养基灌胃;唾液乳杆菌组给予唾液乳杆菌灌胃;甲氨蝶呤组给予甲氨蝶呤药物灌胃,每组连续灌胃4周。结束后评定关节炎指数,测量发病足厚度,观察踝关节组织病理改变并进行组织病理学染色;检测脾脏辅助性T细胞17(Th17)和调节性T细胞(Treg)的含量,评估各组大鼠血清中白细胞介素-1β(IL-1β)、白细胞介素-10(IL-10)、白细胞介素-17(IL-17)、白细胞介素-2(IL-2)、白细胞介素-6(IL-6)、白细胞介素-8(IL-8)、干扰素-γ(IFN-γ)、肿瘤坏死因子-α(TNF-α)的表达水平。结果与健康组相比,CIA模型组、唾液乳杆菌组及甲氨蝶呤组的关节炎指数评分(t=82.31、257.33、234.21),发病足厚度(t=21.47、56.14、51.44),血清中IL-1β(t=33.77、23.52、23.75)、IFN-γ(t=27.89、48.44、56.42)、IL-17(t=51.21、10.50、18.68)、IL-2(t=77.15、11.80、25.16)、IL-6(t=42.96、20.81、18.41)、IL-8(t=64.05、6.47、4.76)、IL-10(t=13.66、2.56、7.88)、TNF-α(t=37.97、30.16、23.07)、Th17(t=61.53、230.11、210.61)水平等均显著升高,差异均有统计学意义(P均<0.05),Treg细胞水平显著下降(t=321.58、2142.17、1820.44),Th17/Treg细胞比值均升高(t=35.12、102.57、123.28),差异均有统计学意义(P均<0.05)。唾液乳杆菌组和甲氨蝶呤组中,脾脏Th17细胞的含量、Th17/Treg比值及关节病理总评分等指标比较,差异均无统计学意义(P均>0.05)。结论唾液乳杆菌对CIA模型SD大鼠具有免疫调节功能和抗炎作用,对应的作用机制主要体现在对Th17和Treg细胞平衡的调节,抑制了IL-1β、IFN-γ、IL-17、IL-2、IL-6、IL-8、IL-10和TNF-α等炎症因
Objective To investigate the therapeutic effect of Lactobacillus salivarius on rheumatoid arthritis(RA).Methods Cattle typeⅡcollagen was used as the immunogen;a collagen induced arthritis(CIA)model was established in male SD rats.Twenty-four successfully modeled rats were randomly divided into CIA model group,Lactobacillus salivarius group,and methotrexate group,with 8 rats in each group.8 rats in the healthy group.Both the healthy group and CIA group were given bacterial culture medium by gavage.The Lactobacillus salivarius group was given Lactobacillus salivarius by gavage.The methotrexate group was given methotrexate medication by gavage,and each group was gavaged continuously for 4 weeks.At the end,the arthritis index was evaluated,the thickness of the affected foot was measured,the pathology of the naked joint tissue,and the histopathological changes were observed.Spleen T helper cell 17(Th17),regulatory T cells(Treg),and the levels of interleukin-1β(IL-1β),interleukin-10(IL-10),interleukin-17(IL-17),interleukin-2(IL-2),interleukin-6(IL-6),interleukin-8(IL-8),interferon-γ(IFN-γ),tumor necrosis factor-α(TNF-α)in serum were determined.Results Compared with the healthy group,the arthritis index score(t=82.31,257.33,234.21),disease foot thickness(t=21.47,56.14,51.44),serum IL-1β(t=33.77,23.52,23.75),IFN-γ(t=27.89,48.44,56.42),IL-17(t=51.21,10.50,18.68),IL-2(t=77.15,11.80,25.16),IL-6(t=42.96,20.81,18.41),IL-8(t=64.05,6.47,4.76),IL-10(t=13.66,2.56,7.88),TNF-α(t=37.97,30.16,23.07),Th17 cells(t=61.53,230.11,210.61)levels in the CIA model group,Lactobacillus salivarius group and methotrexate group were all significantly increased,and the differences were statistically significant(all P<0.05).The level of Treg cells decreased significantly(t=321.58,2142.17,1820.44),the Th17/Treg cell ratio increased in both the Lactobacillus salivarius group and methotrexate group(t=35.12,102.57,123.28),the differences were statistically significant(all P<0.05).There were no statistically significant differences between the
