Aging biomarkers are a combination of biological parameters to(i)assess age-related changes,(ii)track the physiological aging process,and(iii)predict the transition into a pathological status.Although a broad spectrum...Aging biomarkers are a combination of biological parameters to(i)assess age-related changes,(ii)track the physiological aging process,and(iii)predict the transition into a pathological status.Although a broad spectrum of aging biomarkers has been developed,their potential uses and limitations remain poorly characterized.An immediate goal of biomarkers is to help us answer the following three fundamental questions in aging research:How old are we?Why do we get old?And how can we age slower?This review aims to address this need.Here,we summarize our current knowledge of biomarkers developed for cellular,organ,and organismal levels of aging,comprising six pillars:physiological characteristics,medical imaging,histological features,cellular alterations,molecular changes,and secretory factors.To fulfill all these requisites,we propose that aging biomarkers should qualify for being specific,systemic,and clinically relevant.展开更多

Minichromosome Maintenance protein 10(MCM10)is essential for DNA replication initiation and DNA elongation in yeasts and animals.Although the functions of MCM10 in DNA replication and repair have been well documented,...Minichromosome Maintenance protein 10(MCM10)is essential for DNA replication initiation and DNA elongation in yeasts and animals.Although the functions of MCM10 in DNA replication and repair have been well documented,the detailed mechanisms for MCM10 in these processes are not well known.Here,we identified AtMCM10 gene through a forward genetic screening for releasing a silenced marker gene.Although plant MCM10 possesses a similar crystal structure as animal MCM10,AtMCM10 is not essential for plant growth or development in Arabidopsis.AtMCM10 can directly bind to histone H3-H4 and promotes nucleosome assembly in vitro.The nucleosome density is decreased in Atmcm10,and most of the nucleosome density decreased regions in Atmcm10 are also regulated by newly synthesized histone chaperone Chromatin Assembly Factor-1(CAF-1).Loss of both AtMCM10 and CAF-1 is embryo lethal,indicating that AtM CM10 and CAF-1 are indispensable for replication-coupled nucleosome assembly.AtMCM10 interacts with both new and parental histones.Atmcm10 mutants have lower H3.1abundance and reduced H3K27me1/3 levels with releasing some silenced transposons.We propose that AtM CM10 deposits new and parental histones during nucleosome assembly,maintaining proper epigenetic modifications and genome stability during DNA replication.展开更多

Rice mutant 1297 without lipoxygenase isoenzyme-1 (LOX-1) or lipoxygenase isoenzyme-2 (LOX-2) generated by ion beam irradiation from Wanjian2090 and reversion mutant RM1297 with LOX-1 and LOX-2 were subjected to an accelerated-aging experiment. Shanyou63 (with LOX-1 and LOX-2 ) served as control. Results showed that the germination and dehydrogenase activity decreased while the electrical conductivity and free fatty acid content increased in all varieties with accelerated aging. In 1297 that lacked LOX-1 and 2, there were slight changes in germination, dehydrogenase activity, membrane permeability and free fatty acid content during the thirty-day accelerated-aging experiment. But in varieties with LOX-1 and LOX-2, significant changes were observed, suggesting that LOX-1, 2 might be a definite factor which influenced seed lifespan. This study also indicates that ion beam irradiation may be used as mutagen to generate mutant and reversion mutants for biological study and could become a new direction in ion beam application.展开更多
Background:The analysis of chromatin integrity has become an important determinant of sperm quality.In frozenthawed bovine sperm,neither the sequence of post-thaw injury events nor the dynamics of different types of s...Background:The analysis of chromatin integrity has become an important determinant of sperm quality.In frozenthawed bovine sperm,neither the sequence of post-thaw injury events nor the dynamics of different types of sperm DNA breaks are well understood.The aim of the present work was to describe such sperm degradation aftermath focusing on DNA damage dynamics,and to assess if this parameter can predict pregnancy rates in cattle.Results:A total of 75 cryopreserved ejaculates from 25 Holstein bulls were evaluated at two post-thawing periods(0-2 h and 2-4 h),analyzing global and double-stranded DNA damage through alkaline and neutral Comet assays,chromatin deprotamination and decondensation,sperm motility,viability,acrosomal status,and intracellular levels of total ROS,superoxides and calcium.Insemination of 59,605 females was conducted using sperm from the same bulls,thus obtaining the non-return to estrus rates after 90 d(NRR).Results showed an increased rate of double-stranded breaks in the first period(0-2 h:1.29±1.01%/h vs.2-4 h:0.13±1.37%/h;P<0.01),whereas the rate of sperm with moderate+high single-stranded breaks was higher in the second period(0-2 h:3.52±7.77%/h vs.2-4h:21.06±11.69%/h;P<0.0001).Regarding sperm physiology,viability decrease rate was different between the two periods(0-2 h:-4.49±1.79%/h vs.2-4 h:-2.50±3.39%/h;P=0.032),but the progressive motility decrease rate was constant throughout post-thawing incubation(0-2 h:-4.70±3.42%/h vs.2-4 h:-1.89±2.97%/h;P>0.05).Finally,whereas no correlations between bull fertility and any dynamic parameter were found,there were correlations between the NRR and the basal percentage of highly-damaged sperm assessed with the alkaline Comet(Rs=-0.563,P=0.003),between NRR and basal progressive motility(Rs=0.511,P=0.009),and between NRR and sperm with high ROS at 4 h post-thaw(Rs=0.564,P=0.003).Conclusion:The statistically significant correlations found between intracellular ROS,sperm viability,sperm motility,DNA damage and chromatin deprotamination suggested 展开更多

The semolina content in the starting p...Porous Si3N4-SiO2-based ceramics with different porosity were prepared via free sintering of Si3N4 on air with an addition of semolina (5, 10 and 20 wt%) as a pore-forming agent. The semolina content in the starting powder controlled the volume fraction of pores in the sintered body. Small pores (5 μm) formed a continuous network in the whole volume of the ceramic material, while the large pores (~100 μm), formed from the added semolina were mostly isolated in the ceramic matrix. Mercury porosimetry and strength measurements have shown that specific surface area, volume density and compressive strength decreased with the amount of semolina in the samples. Mechanical properties similar to bone were obtained for the sample with 20 wt% semolina pore forming agent (compressive strength 350 MPa, density 2.17 g.cm-3). The prepared Si3N4-SiO2-based ceramics were evaluated for cytotoxic and genotoxic potential on human fibroblast VH10 and B-HNF-1 cells. Biological tests have shown that both these human fibroblast cell lines were sensitive to the samples with lower porosity and cell growth inhibition was observed in the range 14.9% - 21.3%. The cytotoxicity of the sample with the highest porosity (~40%) was not significant (10%). The microscopic observations have shown that VH10 and B-HNF-1 cells growing around the silicon nitride ceramic discs were homogeneously distributed on the cultivation surface. No significant morphologic changes were found in treated cells, their morphology was very similar to that of the control cells. None of the tested Si3N4-based ceramic samples induced necrotic/apoptotic death of human fibroblasts. Sample S-20 had similar properties to bones and was characterized by very good biocompatibility, slight cytotoxicity and none genotoxicity. Therefore, Si3N4-SiO2-based ceramics prepared by free sintering on air are potential biomaterials for medical applications.展开更多
Plants accumulate a vast array of secondary metabolites,which constitute a natural resource for pharmaceuticals.Oldenlandia corymbosa belongs to the Rubiaceae family,and has been used in traditional medicine to treat ...Plants accumulate a vast array of secondary metabolites,which constitute a natural resource for pharmaceuticals.Oldenlandia corymbosa belongs to the Rubiaceae family,and has been used in traditional medicine to treat different diseases,including cancer.However,the active metabolites of the plant,their biosynthetic pathway and mode of action in cancer are unknown.To fill these gaps,we exposed this plant to eight different stress conditions and combined different omics data capturing gene expression,metabolic profiles,and anti-cancer activity.Our results show that O.corymbosa extracts are active against breast cancer cell lines and that ursolic acid is responsible for this activity.Moreover,we assembled a high-quality genome and uncovered two genes involved in the biosynthesis of ursolic acid.Finally,we also revealed that ursolic acid causes mitotic catastrophe in cancer cells and identified three high-confidence protein binding targets by Cellular Thermal Shift Assay(CETSA)and reverse docking.Altogether,these results constitute a valuable resource to further characterize the biosynthesis of active metabolites in the Oldenlandia group,while the mode of action of ursolic acid will allow us to further develop this valuable compound.展开更多

However, the signaling mechanism underlying ethylen...Microtubule reorientation is a long-standing observation that has been implicated in regulating the inhibitory effect of ethylene on axial elongation of plant cells. However, the signaling mechanism underlying ethylene-induced microtubule reorientation has re- mained elusive. Here, we reveal, by live confocal imaging and kinetic root elongation assays, that the time courses of ethylene-induced microtubule reorientation and root elongation inhibition are highly correlated, and that microtubule reorientation is required for the full responsiveness of root elongation to ethylene treatment. Our genetic analysis demonstrated that the effect of ethylene on microtubule orientation and root elongation is mainly transduced through the canonical linear ethylene signaling pathway. By using pharmacological and genetic analyses, we demonstrate further that the TIR1/AFBs-Aux/IAAs-ARFs auxin signaling pathway, but not the ABP1-ROP6-RlC1 auxin signaling branch, is essential for ethylene-induced microtubule reorientation and root elongation inhibition. Together, these findings offer evidence for the functional significance and elucidate the signaling mechanism for ethylene-induced microtubule reorientation in fast root elongation inhibition in Arabidopsis.展开更多
Lysine-ε-acetylation(Kac)is a post-translational modification(PTM)that is critical for metabolic regulation and cell signaling in mammals.However,its prevalence and importance in plants remain to be determined.Employ...Lysine-ε-acetylation(Kac)is a post-translational modification(PTM)that is critical for metabolic regulation and cell signaling in mammals.However,its prevalence and importance in plants remain to be determined.Employing high-resolution tandem mass spectrometry,we analyzed protein lysine acetylation in five representative Arabidopsis organs with 2~3 biological replicates per organ.A total of 2887 Kac proteins and 5929 Kac sites were identified.This comprehensive catalog allows us to analyze proteome-wide features of lysine acetylation.We found that Kac proteins tend to be more uniformly expressed in different organs,and the acetylation status exhibits little correlation with the gene expression level,indicating that acetylation is unlikely caused by stochastic processes.Kac preferentially targets evolutionarily conserved proteins and lysine residues,but only a small percentage of Kac proteins are orthologous between rat and Arabidopsis.A large portion of Kac proteins overlap with proteins modified by other PTMs including ubiquitination,SUMOylation and phosphorylation.Although acetylation,ubiquitination and SUMOylation all modify lysine residues,our analyses show that they rarely target the same sites.In addition,we found that“reader”proteins for acetylation and phosphorylation,i.e.,bromodomain-containing proteins and GRF(General Regulatory Factor)/14-3-3 proteins,are intensively modified by the two PTMs,suggesting that they are main crosstalk nodes between acetylation and phosphorylation signaling.Analyses of GRF6/14-3-3λreveal that the Kac level of GRF6 is decreased under alkaline stress,suggesting that acetylation represses plant alkaline response.Indeed,K56ac of GRF6 inhibits its binding to and subsequent activation of the plasma membrane H+-ATPase AHA2,leading to hypersensitivity to alkaline stress.These results provide valuable resources for protein acetylation studies in plants and reveal that protein acetylation suppresses phosphorylation output by acetylating GRF/14-3-3 proteins.展开更多

In rats, hyperlipidemia was induced by feeding High Fat Diet (Lab diet: Dalda: Coconut oil = 4:3:1) and type 2 diabetes was built inside by injecting Alloxan. The sample N. nucifera leaf powder was added in different percentages with the regular Lab Diet for 21 days feeding. In case of hyperlipidemia developed groups, feeding of Nelumbo nucifera leaf powder at, 30% (70% Lab diet + 30% Sample), 20% (80% Lab diet + 20% Sample) and 10% (90% Lab diet + 10% Sample) showed varied but in a nutshell significant (p Nelumbo nucifera leaf powder for 21 days resulted significant decrease in the blood glucose levels of alloxan-induced diabetic rats. Both the percentages of Nelumbo nucifera leaf powder having 20% with 80% Lab Diet and 10% with 90% Lab Diet significantly (p Nelumbo nucifera powder was found to be exerted to the more prominent effects in lowering the blood glucose level. Nelumbo nucifera leaf powder had 75% efficacy rate in lowering the blood glucose level in comparison to the drug control group treated with glibenclamide which was found with the decreasing capability up to 66%. Although not significant, it, based on the data, can be stated that the sample is endowed with the quality of decreasing capability of body weight. Thus, results of the experimental study reveal that the leaf of Nelumbo nucifera has potent hypoglycemic and hypolipidemic properties.展开更多

The impact of foliar-applied zinc nanoparticles (ZnNPs) and combined zinc nano-loaded with moringa extracts (ZnONPs) on salt tolerance in faba beans (cultivar, Giza-716) grown under saline soil (50 and 100 mM NaCl) was investigated. Moringa oleifera extract has been used as a chelating agent to synthesize zinc oxide nanoparticles. The crystalline structure, morphology, and chemical composition of ZnO nanoparticles were studied using various characterization techniques, including UV-visible spectroscopy (UV), Fourier Transform Infrared Analysis (FTIR), scanning electron microscopy (SEM), and X-ray diffraction (XRD). Morphological, chemical, and biochemical parameters of plants at 60 and 90 days after sowing were assessed. Salinity stress caused a remarkable reduction in growth traits, photosynthetic pigments and proline levels of the faba bean. Foliar spray with ZnNPs and ZnONPs on faba bean grown under saline soils promoted plant growth parameters (i.e., shoot length, numbers of leaves, relative water content, shoot and roots fresh and dry weights), photosynthetic pigments (Chl a, b, total chlorophyll, and carotenoids), proline and mineral elements (Na<sup>+</sup>, K<sup>+</sup>, Ca<sup>2+</sup>, and Zn<sup>2+</sup>) compared to control. However, at 100 mM NaCl, there were no significant variations in the mentioned parameters. This study suggested that there is potential for foliar spraying with ZnNPs and ZnONPs in improving growth parameters, photosynthesis efficiency and biochemical aspects of faba bean plants under saline conditions.展开更多

A sum of fourteen different types of spices at different percentages were taken into consideration to propel the research activities forward, viz;ginger, garlic, onion, red chili, turmeric, cumin, cinnamon, clove, coriander, fenugreek, black pepper, nigella, carom seed and cardamom. The healthy experimental wistar albino rats were made diabetic by injecting alloxan monohydrate intraperitoneally a dose of 150 mg/kg body weight. The rats of Group-D and Group-E were treated with the sample, mixed spices at a dose of 100 mg/kg and 200 mg/kg body weight respectively with the lab diet while in case of Group-C, glibenclamide at a dose of 0.5 mg/kg body weight was introduced with the lab diet for a total period of 21 days where as for the same days span, Group-A and Group-B were continued feeding with only lab diet. Data suggest that the mixed spices of both doses applied in Group-D and Group-E had exerted effects significantly (p 0.05) in lowering the blood glucose level. On 22nd day, in Group-D and Group-E, the FBG level was found to be reduced by around 40.66% and 41.18% respectively compared to their respective initial days where in Group-C, the reduction was around 51.90% in comparison to its initial day. Again, in case of 2hPG level measurement, on 22nd day, both the groups, Group-D and Group-E were found with the decreased level of around 19.38% and 39.11% respectively compared to their respective initial days while the Group-C was shown with the reduced of 44.26% in comparison adopting same manner. Furthermore, various analyses confirm the presence of several micro and macro nutrients and high amount of dietary fiber in the spices mix. Thus the result affirms that this combined spice possesses strong hypoglycemic properties.展开更多

Methods: Coriander seeds-and leaves-based sauces were administrated to normal(Study Ⅰ) and carbon tetrachloride(2 mL/kg B.W.)-induced hepatotoxic rabbits(Study Ⅱ). Hepatic and renal biomarkers like aspartate aminotransferase,alanine aminotransferase. alkaline phosphatase, superoxide dismutase and catalase were measured. Results: Coriander leaves-based sauce exerted more decline(P<0.05) in serum aspartate aminotransferase, alanine aminotransferase and alkaline phosphatase levels in the study Ⅱ as 18.32%. 19.91% and 21.24%, respectively. While, hepatic superoxide dismutase and catalase levels were raised significantly(P<0.05) in both studies. Renal parameters also depicted positive impact by the provision of developed sauces. Conclusions: Coriander seeds and leaves based sauces are effective in alleviating the hepato/renal toxicity. The hepatoprotective effect of coriander leaves is more pronounced as compared to coriander seeds.展开更多
Objective:To evaluate the effects of ethanol–water(80:20) extract of Marrubium vulgare(M.vulgare) on the hematological parameters,macroscopic and histological aspects of the uterus and fetus in non-pregnant and pregn...Objective:To evaluate the effects of ethanol–water(80:20) extract of Marrubium vulgare(M.vulgare) on the hematological parameters,macroscopic and histological aspects of the uterus and fetus in non-pregnant and pregnant rats.Methods:Female rats were divided into 4 equal groups(n = 9),group N(normal rats) and group G(pregnant rats) considered as control groups,group NE(normal rats treated with the ethanol–water(80:20,v/v) extract of M.vulgare) and group GE(pregnant rats treated with the extract).The ethanol–water(80:20) plant extract was administered in a single daily dose 1 g/kg at the morning,during 19 d.On the 19 day of the experiment,animals were sacrificed,the uterus and fetuses were removed for the morphological and histological studies and the blood was collected in EDTA tubes for the measurement of hematological parameters with the use of an automate ‘HORIBA ABX Micros 60 Hematology Analyzer'.Results:Our results showed,in group NE and GE,a significant decrease on hematological parameters:red blood cells(NE:18.6%;GE:38.4%),hematocrit(NE:13.8%;GE:20.4%),hemoglobin(NE:12.1%;GE:8.3%) and mean corpuscular volume(NE:6.4%;GE:2%) with P more less a 0.05.Indeed,the extract of M.vulgare caused a significant decrease on the mean implantations of fetuses(82.5%,P<0.001) and their size(47.2%,P< 0.01).As for the macroscopic and histological appearance of uterus,our data showed no change in normal treated rats.In contrast,the treated pregnant rats showed a severe histological change characterized by the existence of location of stopped gestation.Furthermore,it was also found in the uterus of these rat lyses placental and embryo tissue.Conclusions:All these results support the hypothesis of an abortifacient effect of M.vulgare.展开更多

文摘Aging biomarkers are a combination of biological parameters to(i)assess age-related changes,(ii)track the physiological aging process,and(iii)predict the transition into a pathological status.Although a broad spectrum of aging biomarkers has been developed,their potential uses and limitations remain poorly characterized.An immediate goal of biomarkers is to help us answer the following three fundamental questions in aging research:How old are we?Why do we get old?And how can we age slower?This review aims to address this need.Here,we summarize our current knowledge of biomarkers developed for cellular,organ,and organismal levels of aging,comprising six pillars:physiological characteristics,medical imaging,histological features,cellular alterations,molecular changes,and secretory factors.To fulfill all these requisites,we propose that aging biomarkers should qualify for being specific,systemic,and clinically relevant.

文摘Minichromosome Maintenance protein 10(MCM10)is essential for DNA replication initiation and DNA elongation in yeasts and animals.Although the functions of MCM10 in DNA replication and repair have been well documented,the detailed mechanisms for MCM10 in these processes are not well known.Here,we identified AtMCM10 gene through a forward genetic screening for releasing a silenced marker gene.Although plant MCM10 possesses a similar crystal structure as animal MCM10,AtMCM10 is not essential for plant growth or development in Arabidopsis.AtMCM10 can directly bind to histone H3-H4 and promotes nucleosome assembly in vitro.The nucleosome density is decreased in Atmcm10,and most of the nucleosome density decreased regions in Atmcm10 are also regulated by newly synthesized histone chaperone Chromatin Assembly Factor-1(CAF-1).Loss of both AtMCM10 and CAF-1 is embryo lethal,indicating that AtM CM10 and CAF-1 are indispensable for replication-coupled nucleosome assembly.AtMCM10 interacts with both new and parental histones.Atmcm10 mutants have lower H3.1abundance and reduced H3K27me1/3 levels with releasing some silenced transposons.We propose that AtM CM10 deposits new and parental histones during nucleosome assembly,maintaining proper epigenetic modifications and genome stability during DNA replication.

文摘Mushrooms are experiencing a kind of renaissance as a part of the contemporary human diet.These valuable organisms are more than food,they fit in perfectly as a novel market group known as nutra-mycoceuticals.Immune-balancing mushroom dietary fibers and secondary metabolites such as polyphenols are the main focus of the healthcare industry.Wellness and cosmetic companies are increasingly using mushroom extracts rich in these ingredients.This review considers the basic molecular immunomodulatory mechanisms of action of the most commonly used mushroom dietary fibers,β-glucans.The literature data on their bioavailability,metabolic transformations,preclinical and human clinical research,and safety are discussed.Immunomodulatory mechanisms of polyphenol ingredients are also considered.These molecules present great potential in the design of the new immunity balancer formulations according to their widespread structural diversity.Finally,we draw attention to the perspectives of modern trends in mushroom nutraceutical and cosmeceutical formulations to strengthen and balance immunity.

文摘The effect of absence of lipoxygenase isoenzyme (LOX) on storage stability was investigated. Rice mutant 1297 without lipoxygenase isoenzyme-1 (LOX-1) or lipoxygenase isoenzyme-2 (LOX-2) generated by ion beam irradiation from Wanjian2090 and reversion mutant RM1297 with LOX-1 and LOX-2 were subjected to an accelerated-aging experiment. Shanyou63 (with LOX-1 and LOX-2 ) served as control. Results showed that the germination and dehydrogenase activity decreased while the electrical conductivity and free fatty acid content increased in all varieties with accelerated aging. In 1297 that lacked LOX-1 and 2, there were slight changes in germination, dehydrogenase activity, membrane permeability and free fatty acid content during the thirty-day accelerated-aging experiment. But in varieties with LOX-1 and LOX-2, significant changes were observed, suggesting that LOX-1, 2 might be a definite factor which influenced seed lifespan. This study also indicates that ion beam irradiation may be used as mutagen to generate mutant and reversion mutants for biological study and could become a new direction in ion beam application.

文摘Background:The analysis of chromatin integrity has become an important determinant of sperm quality.In frozenthawed bovine sperm,neither the sequence of post-thaw injury events nor the dynamics of different types of sperm DNA breaks are well understood.The aim of the present work was to describe such sperm degradation aftermath focusing on DNA damage dynamics,and to assess if this parameter can predict pregnancy rates in cattle.Results:A total of 75 cryopreserved ejaculates from 25 Holstein bulls were evaluated at two post-thawing periods(0-2 h and 2-4 h),analyzing global and double-stranded DNA damage through alkaline and neutral Comet assays,chromatin deprotamination and decondensation,sperm motility,viability,acrosomal status,and intracellular levels of total ROS,superoxides and calcium.Insemination of 59,605 females was conducted using sperm from the same bulls,thus obtaining the non-return to estrus rates after 90 d(NRR).Results showed an increased rate of double-stranded breaks in the first period(0-2 h:1.29±1.01%/h vs.2-4 h:0.13±1.37%/h;P<0.01),whereas the rate of sperm with moderate+high single-stranded breaks was higher in the second period(0-2 h:3.52±7.77%/h vs.2-4h:21.06±11.69%/h;P<0.0001).Regarding sperm physiology,viability decrease rate was different between the two periods(0-2 h:-4.49±1.79%/h vs.2-4 h:-2.50±3.39%/h;P=0.032),but the progressive motility decrease rate was constant throughout post-thawing incubation(0-2 h:-4.70±3.42%/h vs.2-4 h:-1.89±2.97%/h;P>0.05).Finally,whereas no correlations between bull fertility and any dynamic parameter were found,there were correlations between the NRR and the basal percentage of highly-damaged sperm assessed with the alkaline Comet(Rs=-0.563,P=0.003),between NRR and basal progressive motility(Rs=0.511,P=0.009),and between NRR and sperm with high ROS at 4 h post-thaw(Rs=0.564,P=0.003).Conclusion:The statistically significant correlations found between intracellular ROS,sperm viability,sperm motility,DNA damage and chromatin deprotamination suggested

文摘Porous Si3N4-SiO2-based ceramics with different porosity were prepared via free sintering of Si3N4 on air with an addition of semolina (5, 10 and 20 wt%) as a pore-forming agent. The semolina content in the starting powder controlled the volume fraction of pores in the sintered body. Small pores (5 μm) formed a continuous network in the whole volume of the ceramic material, while the large pores (~100 μm), formed from the added semolina were mostly isolated in the ceramic matrix. Mercury porosimetry and strength measurements have shown that specific surface area, volume density and compressive strength decreased with the amount of semolina in the samples. Mechanical properties similar to bone were obtained for the sample with 20 wt% semolina pore forming agent (compressive strength 350 MPa, density 2.17 g.cm-3). The prepared Si3N4-SiO2-based ceramics were evaluated for cytotoxic and genotoxic potential on human fibroblast VH10 and B-HNF-1 cells. Biological tests have shown that both these human fibroblast cell lines were sensitive to the samples with lower porosity and cell growth inhibition was observed in the range 14.9% - 21.3%. The cytotoxicity of the sample with the highest porosity (~40%) was not significant (10%). The microscopic observations have shown that VH10 and B-HNF-1 cells growing around the silicon nitride ceramic discs were homogeneously distributed on the cultivation surface. No significant morphologic changes were found in treated cells, their morphology was very similar to that of the control cells. None of the tested Si3N4-based ceramic samples induced necrotic/apoptotic death of human fibroblasts. Sample S-20 had similar properties to bones and was characterized by very good biocompatibility, slight cytotoxicity and none genotoxicity. Therefore, Si3N4-SiO2-based ceramics prepared by free sintering on air are potential biomaterials for medical applications.

文摘Plants accumulate a vast array of secondary metabolites,which constitute a natural resource for pharmaceuticals.Oldenlandia corymbosa belongs to the Rubiaceae family,and has been used in traditional medicine to treat different diseases,including cancer.However,the active metabolites of the plant,their biosynthetic pathway and mode of action in cancer are unknown.To fill these gaps,we exposed this plant to eight different stress conditions and combined different omics data capturing gene expression,metabolic profiles,and anti-cancer activity.Our results show that O.corymbosa extracts are active against breast cancer cell lines and that ursolic acid is responsible for this activity.Moreover,we assembled a high-quality genome and uncovered two genes involved in the biosynthesis of ursolic acid.Finally,we also revealed that ursolic acid causes mitotic catastrophe in cancer cells and identified three high-confidence protein binding targets by Cellular Thermal Shift Assay(CETSA)and reverse docking.Altogether,these results constitute a valuable resource to further characterize the biosynthesis of active metabolites in the Oldenlandia group,while the mode of action of ursolic acid will allow us to further develop this valuable compound.

文摘Microtubule reorientation is a long-standing observation that has been implicated in regulating the inhibitory effect of ethylene on axial elongation of plant cells. However, the signaling mechanism underlying ethylene-induced microtubule reorientation has re- mained elusive. Here, we reveal, by live confocal imaging and kinetic root elongation assays, that the time courses of ethylene-induced microtubule reorientation and root elongation inhibition are highly correlated, and that microtubule reorientation is required for the full responsiveness of root elongation to ethylene treatment. Our genetic analysis demonstrated that the effect of ethylene on microtubule orientation and root elongation is mainly transduced through the canonical linear ethylene signaling pathway. By using pharmacological and genetic analyses, we demonstrate further that the TIR1/AFBs-Aux/IAAs-ARFs auxin signaling pathway, but not the ABP1-ROP6-RlC1 auxin signaling branch, is essential for ethylene-induced microtubule reorientation and root elongation inhibition. Together, these findings offer evidence for the functional significance and elucidate the signaling mechanism for ethylene-induced microtubule reorientation in fast root elongation inhibition in Arabidopsis.

文摘Lysine-ε-acetylation(Kac)is a post-translational modification(PTM)that is critical for metabolic regulation and cell signaling in mammals.However,its prevalence and importance in plants remain to be determined.Employing high-resolution tandem mass spectrometry,we analyzed protein lysine acetylation in five representative Arabidopsis organs with 2~3 biological replicates per organ.A total of 2887 Kac proteins and 5929 Kac sites were identified.This comprehensive catalog allows us to analyze proteome-wide features of lysine acetylation.We found that Kac proteins tend to be more uniformly expressed in different organs,and the acetylation status exhibits little correlation with the gene expression level,indicating that acetylation is unlikely caused by stochastic processes.Kac preferentially targets evolutionarily conserved proteins and lysine residues,but only a small percentage of Kac proteins are orthologous between rat and Arabidopsis.A large portion of Kac proteins overlap with proteins modified by other PTMs including ubiquitination,SUMOylation and phosphorylation.Although acetylation,ubiquitination and SUMOylation all modify lysine residues,our analyses show that they rarely target the same sites.In addition,we found that“reader”proteins for acetylation and phosphorylation,i.e.,bromodomain-containing proteins and GRF(General Regulatory Factor)/14-3-3 proteins,are intensively modified by the two PTMs,suggesting that they are main crosstalk nodes between acetylation and phosphorylation signaling.Analyses of GRF6/14-3-3λreveal that the Kac level of GRF6 is decreased under alkaline stress,suggesting that acetylation represses plant alkaline response.Indeed,K56ac of GRF6 inhibits its binding to and subsequent activation of the plasma membrane H+-ATPase AHA2,leading to hypersensitivity to alkaline stress.These results provide valuable resources for protein acetylation studies in plants and reveal that protein acetylation suppresses phosphorylation output by acetylating GRF/14-3-3 proteins.
文摘The study was carried out to evaluate the hypolipidemic and hypoglycemic effect of Nelumbo nucifera leaf powder. In rats, hyperlipidemia was induced by feeding High Fat Diet (Lab diet: Dalda: Coconut oil = 4:3:1) and type 2 diabetes was built inside by injecting Alloxan. The sample N. nucifera leaf powder was added in different percentages with the regular Lab Diet for 21 days feeding. In case of hyperlipidemia developed groups, feeding of Nelumbo nucifera leaf powder at, 30% (70% Lab diet + 30% Sample), 20% (80% Lab diet + 20% Sample) and 10% (90% Lab diet + 10% Sample) showed varied but in a nutshell significant (p Nelumbo nucifera leaf powder for 21 days resulted significant decrease in the blood glucose levels of alloxan-induced diabetic rats. Both the percentages of Nelumbo nucifera leaf powder having 20% with 80% Lab Diet and 10% with 90% Lab Diet significantly (p Nelumbo nucifera powder was found to be exerted to the more prominent effects in lowering the blood glucose level. Nelumbo nucifera leaf powder had 75% efficacy rate in lowering the blood glucose level in comparison to the drug control group treated with glibenclamide which was found with the decreasing capability up to 66%. Although not significant, it, based on the data, can be stated that the sample is endowed with the quality of decreasing capability of body weight. Thus, results of the experimental study reveal that the leaf of Nelumbo nucifera has potent hypoglycemic and hypolipidemic properties.
文摘Salinity stress limits crop growth and productivity, including legumes in various regions worldwide. The impact of foliar-applied zinc nanoparticles (ZnNPs) and combined zinc nano-loaded with moringa extracts (ZnONPs) on salt tolerance in faba beans (cultivar, Giza-716) grown under saline soil (50 and 100 mM NaCl) was investigated. Moringa oleifera extract has been used as a chelating agent to synthesize zinc oxide nanoparticles. The crystalline structure, morphology, and chemical composition of ZnO nanoparticles were studied using various characterization techniques, including UV-visible spectroscopy (UV), Fourier Transform Infrared Analysis (FTIR), scanning electron microscopy (SEM), and X-ray diffraction (XRD). Morphological, chemical, and biochemical parameters of plants at 60 and 90 days after sowing were assessed. Salinity stress caused a remarkable reduction in growth traits, photosynthetic pigments and proline levels of the faba bean. Foliar spray with ZnNPs and ZnONPs on faba bean grown under saline soils promoted plant growth parameters (i.e., shoot length, numbers of leaves, relative water content, shoot and roots fresh and dry weights), photosynthetic pigments (Chl a, b, total chlorophyll, and carotenoids), proline and mineral elements (Na<sup>+</sup>, K<sup>+</sup>, Ca<sup>2+</sup>, and Zn<sup>2+</sup>) compared to control. However, at 100 mM NaCl, there were no significant variations in the mentioned parameters. This study suggested that there is potential for foliar spraying with ZnNPs and ZnONPs in improving growth parameters, photosynthesis efficiency and biochemical aspects of faba bean plants under saline conditions.
文摘The main purpose of the study was to reveal the hypoglycemic effect of a combination of some selected spices along with the evaluation of nutritional values. A sum of fourteen different types of spices at different percentages were taken into consideration to propel the research activities forward, viz;ginger, garlic, onion, red chili, turmeric, cumin, cinnamon, clove, coriander, fenugreek, black pepper, nigella, carom seed and cardamom. The healthy experimental wistar albino rats were made diabetic by injecting alloxan monohydrate intraperitoneally a dose of 150 mg/kg body weight. The rats of Group-D and Group-E were treated with the sample, mixed spices at a dose of 100 mg/kg and 200 mg/kg body weight respectively with the lab diet while in case of Group-C, glibenclamide at a dose of 0.5 mg/kg body weight was introduced with the lab diet for a total period of 21 days where as for the same days span, Group-A and Group-B were continued feeding with only lab diet. Data suggest that the mixed spices of both doses applied in Group-D and Group-E had exerted effects significantly (p 0.05) in lowering the blood glucose level. On 22nd day, in Group-D and Group-E, the FBG level was found to be reduced by around 40.66% and 41.18% respectively compared to their respective initial days where in Group-C, the reduction was around 51.90% in comparison to its initial day. Again, in case of 2hPG level measurement, on 22nd day, both the groups, Group-D and Group-E were found with the decreased level of around 19.38% and 39.11% respectively compared to their respective initial days while the Group-C was shown with the reduced of 44.26% in comparison adopting same manner. Furthermore, various analyses confirm the presence of several micro and macro nutrients and high amount of dietary fiber in the spices mix. Thus the result affirms that this combined spice possesses strong hypoglycemic properties.

文摘Objective: To evaluate the hepatic and renal protective potential of coriander seeds and leaves using animal feed model. Methods: Coriander seeds-and leaves-based sauces were administrated to normal(Study Ⅰ) and carbon tetrachloride(2 mL/kg B.W.)-induced hepatotoxic rabbits(Study Ⅱ). Hepatic and renal biomarkers like aspartate aminotransferase,alanine aminotransferase. alkaline phosphatase, superoxide dismutase and catalase were measured. Results: Coriander leaves-based sauce exerted more decline(P<0.05) in serum aspartate aminotransferase, alanine aminotransferase and alkaline phosphatase levels in the study Ⅱ as 18.32%. 19.91% and 21.24%, respectively. While, hepatic superoxide dismutase and catalase levels were raised significantly(P<0.05) in both studies. Renal parameters also depicted positive impact by the provision of developed sauces. Conclusions: Coriander seeds and leaves based sauces are effective in alleviating the hepato/renal toxicity. The hepatoprotective effect of coriander leaves is more pronounced as compared to coriander seeds.

文摘Objective:To evaluate the effects of ethanol–water(80:20) extract of Marrubium vulgare(M.vulgare) on the hematological parameters,macroscopic and histological aspects of the uterus and fetus in non-pregnant and pregnant rats.Methods:Female rats were divided into 4 equal groups(n = 9),group N(normal rats) and group G(pregnant rats) considered as control groups,group NE(normal rats treated with the ethanol–water(80:20,v/v) extract of M.vulgare) and group GE(pregnant rats treated with the extract).The ethanol–water(80:20) plant extract was administered in a single daily dose 1 g/kg at the morning,during 19 d.On the 19 day of the experiment,animals were sacrificed,the uterus and fetuses were removed for the morphological and histological studies and the blood was collected in EDTA tubes for the measurement of hematological parameters with the use of an automate ‘HORIBA ABX Micros 60 Hematology Analyzer'.Results:Our results showed,in group NE and GE,a significant decrease on hematological parameters:red blood cells(NE:18.6%;GE:38.4%),hematocrit(NE:13.8%;GE:20.4%),hemoglobin(NE:12.1%;GE:8.3%) and mean corpuscular volume(NE:6.4%;GE:2%) with P more less a 0.05.Indeed,the extract of M.vulgare caused a significant decrease on the mean implantations of fetuses(82.5%,P<0.001) and their size(47.2%,P< 0.01).As for the macroscopic and histological appearance of uterus,our data showed no change in normal treated rats.In contrast,the treated pregnant rats showed a severe histological change characterized by the existence of location of stopped gestation.Furthermore,it was also found in the uterus of these rat lyses placental and embryo tissue.Conclusions:All these results support the hypothesis of an abortifacient effect of M.vulgare.
